Condition-dependent structural collapse in the intrinsically disordered N-terminal domain of prion protein

Summary: The study revealed that the conformation and size of the prion protein are sensitive to solvent conditions. The N-domain and C-domain exhibited different folding and aggregation tendencies under low salt concentration and acidic conditions, respectively. Positive residues in the N-domain played a role in protecting the exposed hydrophobic surface of the C-domain.

Location of the cross-β structure in prion fibrils: A search by seeding and electron spin resonance spectroscopy

Summary: This study used a combination of site-directed nitroxide spin-labeling, fibril seeding, and electron spin resonance (ESR) spectroscopy to determine the structure of in vitro-prepared full-length mouse prion fibrils. The formation of the amyloid core was found to be linked to an alpha-to-beta structural transformation involving the segment 160-224. This method is suitable for examining the structure of hetero-seeded amyloid fibrils.

Structural basis for the helical filament formation of Escherichia coli glutamine synthetase

Summary: This study reveals the structure of the metal ion-induced filament of Escherichia coli glutamine synthetase (EcGS) using cryo-electron microscopy. It was found that the filament formation of EcGS involves stacking of native dodecamers through chelating nickel ions. However, upon adding glutamate or ATP for catalytic reactions, the filament reverts back to individual dodecamers.

Aggregation Condition-Structure Relationship of Mouse Prion Protein Fibrils

Summary: Researchers have successfully developed structural models of mouse prion protein fibrils prepared under three different denaturing conditions using atomic force microscopy, FTIR spectroscopy, and magic-angle spinning NMR. The study revealed that while the core region and N- and C-terminal structures are almost identical among the three fibrils, the central part differs in terms of beta-strand length and charged residue arrangement. This suggests that the ionic strength of the denaturant plays a key role in determining the structure of fibrils under specific conditions.
