Identification and Functional Analysis of Cytokine-Like Protein CLEC-47 in Caenorhabditis elegans

Summary: Effector proteins in Caenorhabditis elegans play a role in host defense, and this study focused on identifying pathogen-dependent cytokine-like molecules, specifically C-type lectin domain-containing proteins (CLECs). The results show that CLEC-47 functions as a novel cytokine-like signaling molecule, attracting and recruiting macrophages and activating proinflammatory cytokine expression.

Isobaric Peptide Labeling on Digital Microfluidics for Quantitative Low Cell Number Proteomics

Summary: Digital microfluidics (DMF) is a technology suitable for miniaturized, automated, and multiplexed liquid handling in bioanalytical applications. While its use in LC-MS-based proteomics has been limited to qualitative analyses, the implementation of isobaric peptide labeling on DMF devices has shown promise, especially with the use of maltoside-based detergents. This detergent enables facile droplet movement and is compatible with organic solvents, allowing for efficient isobaric tandem mass tag (TMT) labeling. The newly developed labeling protocol has been successfully applied to multiplexed proteome studies, showing promise for high throughput low cell number proteomics.

Perfluorooctane sulfonate (PFOS) disturbs fatty acid metabolism in Caenorhabditis elegans: Evidence from chemical analysis and molecular mechanism exploration

Summary: This study revealed that exposure to PFOS led to disturbances in fatty acid metabolism in C. elegans, characterized by decreased levels of C18:0, increased levels of C18:3n6, decreased C18:3n6: C18:2n6 ratio, and down-regulation of the FA desaturation gene fat-3. Additionally, exposure disrupted mitochondrial function, leading to reduced ATP synthesis.

Ultrasensitive single-cell proteomics workflow identifies >1000 protein groups per mammalian cell

Summary: The combination of nanodroplet sample preparation, ultra-low-flow nanoLC, high-field asymmetric ion mobility spectrometry (FAIMS), and the latest-generation Orbitrap Eclipse Tribrid mass spectrometer led to greatly improved single-cell proteome profiling. FAIMS effectively filtered out singly charged ions, allowing for more effective MS analysis of multiply charged peptides, resulting in a significantly higher number of protein groups identified from single HeLa cells compared to traditional methods. Differential analysis also showed similar proteome coverage for single microdissected motor neurons and interneurons from human spinal tissue, with the ability to differentiate between the two subpopulations based on label-free quantification.
