Signaling Mechanisms and Pharmacological Modulators Governing Diverse Aquaporin Functions in Human Health and Disease

Kim Wagner et al.

Summary: This review highlights the key roles of aquaporins (AQPs) in fluid homeostasis, glandular secretions, signal transduction and sensation, barrier function, immunity and inflammation, cell migration, and angiogenesis. It emphasizes that the functions of AQPs are more complex than simply mediating the flow of water. Further research and development of AQP inhibitors will provide new insights and treatments for AQP-associated disorders.

The effects of trifluoperazine on brain edema, aquaporin-4 expression and metabolic markers during the acute phase of stroke using photothrombotic mouse model

Nicole J. Sylvain et al.

Summary: Stroke is the second leading cause of death and the third leading cause of disability worldwide, with edema playing a major role in stroke-associated morbidity and mortality. Regulating water balance in the brain through targeting AQP4 channels may be a promising therapeutic approach for treating brain edema. The FDA-approved drug trifluoperazine (TFP) has shown efficacy in reducing cerebral edema in post-stroke mice by inhibiting AQP4 expression, without inducing deleterious changes in brain electrolytes or metabolic markers.

Direct imaging of glymphatic transport using H217O MRI

Mohammed S. Alshuhri et al.

Summary: The recently proposed glymphatic pathway for solute transport and waste clearance from the brain has been the focus of intense debate. By using an isotopically enriched MRI tracer, researchers directly imaged glymphatic water transport in the rat brain in vivo, revealing faster and more extensive transport than previously thought, with aquaporin-4 channels playing a critical role.

CXCL12-mediated monocyte transmigration into brain perivascular space leads to neuroinflammation and memory deficit in neuropathic pain

Chun-Lin Mai et al.

Summary: The study reveals that in cognitive impairment caused by neuroinflammation, the recruitment of monocytes into the perivascular space mediated by CXCL12 plays a critical role, and blocking this process can alleviate memory decline caused by neuropathic pain.
