Functional Characterization and Protein Engineering of a Triterpene 3-/6-/2′-O-Glycosyltransferase Reveal a Conserved Residue Critical for the Regiospecificity

Summary: A multifunctional glycosyltransferase AmGT8 was identified for catalyzing triterpene glycosylation from Astragalus membranaceus. Through a small mutant library, regiospecificity of AmGT8 was successfully engineered, leading to the synthesis of astragalosides III and IV. This work provides new insights into protein engineering of regioselectivity in plant GTs.

Functional and Structural Dissection of a Plant Steroid 3-O-Glycosyltransferase Facilitated the Engineering Enhancement of Sugar Donor Promiscuity

Summary: Cardiotonic steroids (CTSs) are important drugs for treating heart failure. Glycosylation of CTSs plays a significant role in their pharmacological applications. Plant steroid 3-O-glycosyltransferase (S3GT) is a tool to achieve CTS glycosylation, but a S3GT with both substrate and sugar donor promiscuity is still unidentified. This study identified and characterized a plant S3GT, UGT74AN2, and revealed the molecular basis for CTS glycosylation. Additionally, an engineered variant of UGT74AN2 with enhanced sugar donor promiscuity was constructed.

Promiscuity Characteristics of Versatile Plant Glycosyltransferases for Natural Product Glycodiversification

Summary: This study systematically investigates the relationship between phylogeny and substrate promiscuity in plant GT1-pGTs. By analyzing available sequences and conducting experiments on selected GTs, the researchers identify highly promiscuous GTs that have a wide tolerance for different substrates and can catalyze the formation of different types of glycosidic bonds. The findings contribute to the discovery of more promiscuous plant GTs and enhance the enzymatic toolkit for sustainable production of valuable glycosides.

Protein Engineering of PhUGT, a Donor Promiscuous Glycosyltransferase, for the Improved Enzymatic Synthesis of Antioxidant Quercetin 3-O-N-Acetylgalactosamine

Summary: In this study, Quercetin 3-O-N-acetylgalactosamine (Q3GalNAc), a compound that had never been enzymatically synthesized before, was successfully produced. The enzyme PhUGT was identified to have quercetin GalNAc-T activity and was engineered to enhance the conversion rate. The study also revealed the donor promiscuity of PhUGT and provided a potent biocatalyst for glycodiversifying quercetin.
