Development of a Highly Selective Plasmodium falciparum Proteasome Inhibitor with Anti-malaria Activity in Humanized Mice

Summary: Studies have shown that Plasmodium falciparum proteasome inhibitors have the potential to treat, prevent, and block transmission of malaria. A noncovalent, macrocyclic peptide inhibitor with high species selectivity and improved pharmacokinetic properties has been developed, demonstrating specific inhibition of the Pf20S beta 5 subunit.

Microbial proteasomes as drug targets

Summary: Proteasomes are ATP-dependent, N-terminal nucleophile hydrolases that are essential for intracellular protein turnover and present in all biological kingdoms. Inhibiting proteasomes can be harmful to cell viability, but proteasome inhibitor drugs have revolutionized the treatment of multiple myeloma. Efforts are being made to develop inhibitors selective for microbial proteasomes, with promising progress in targeting pathogenic microbes for diseases like leishmaniasis and Chagas disease.

Macrocyclic Peptides that Selectively Inhibit the Mycobacterium tuberculosis Proteasome

Summary: Current treatment of tuberculosis (TB) typically lasts at least 6 months, with the latent Mycobacterium tuberculosis (Mtb) being tolerant to most anti-TB drugs. A key hypothesis is that drugs targeting the Mtb proteasome (Mtb20S) may shorten treatment time when used in combination with conventional drugs. A series of macrocyclic peptides, including macrocycle 6, have been developed that selectively target the Mtb20S and have the potential to be developed as anti-TB therapeutics.
