Multiomic profiling of checkpoint inhibitor-treated melanoma: Identifying predictors of response and resistance, and markers of biological discordance

Felicity Newell et al.

Summary: This study examines the relationship between various factors and the response to immunotherapy in patients with advanced cutaneous melanoma. The results identify several factors associated with response and develop a multivariable model for predicting response. However, the study also indicates the presence of intratumor and intertumor heterogeneity in response to immunotherapy and the lack of clear biological mechanisms explaining non-predicted responses.

Comparative investigation of cell cycle and immunomodulatory genes in mucosal and cutaneous melanomas: Preliminary data suggest a potential promising clinical role for p16 and the PD-1/PD-L1 axis

Niklas Wrede et al.

Summary: Mucosal melanomas and cutaneous melanomas exhibit different levels of expression of cell cycle-associated and immunomodulatory proteins, which are partially regulated by DNA methylation and copy number alterations. PD-1 expression and PD-L1 promoter methylation levels may serve as prognostic markers for mucosal melanomas.

Tumor Intrinsic PD-L1 Promotes DNA Repair in Distinct Cancers and Suppresses PARP Inhibitor-Induced Synthetic Lethality

Anand V. R. Kornepati et al.

Summary: BRCA1-mediated homologous recombination is regulated by the immune checkpoint molecule PD-L1, with genetic depletion of tumor PD-L1 affecting DNA repair mechanisms and sensitizing tumors to PARP inhibitors, highlighting a potential therapeutic vulnerability and response biomarker.

CTLA4 promoter hypomethylation is a negative prognostic biomarker at initial diagnosis but predicts response and favorable outcome to anti-PD-1 based immunotherapy in clear cell renal cell carcinoma

Niklas Kluemper et al.

Summary: The study found that CTLA4 promoter hypomethylation is associated with CTLA4 mRNA expression, lymphocyte infiltration, and poor prognosis in ccRCC. However, in patients with ICB-treated ccRCC, CTLA4 promoter hypomethylation predicted treatment response and favorable outcomes. Therefore, CTLA4 methylation may serve as a powerful predictive biomarker for immunotherapy response in metastatic RCC.
