The two-stage interaction of Ebola virus VP40 with nucleoprotein results in a switch from viral RNA synthesis to virion assembly/budding

Summary: A study found that the N-terminus of the Ebola virus nucleoprotein contributes to the release of virus-like particles (VLPs) containing nucleoprotein by recruiting the matrix protein VP40. Mutations in the C-terminal hydrophobic core of nucleoprotein strengthen the interaction with VP40, leading to reduced VLP release and inhibition of viral RNA synthesis. This suggests that the interaction between the N-terminus of nucleoprotein and VP40 induces a conformational change in the C-terminus of nucleoprotein, inhibiting RNA synthesis and initiation of virion assembly/budding.

Summary: This study elucidated the mechanism by which M95 affects cell death induced by rabies virus infection. Ni-CE strain induced DNA fragmentation, cell membrane disruption, PS exposure, caspase-3/7 activation, and PARP-1 cleavage, while Nishigahara strain induced PS exposure but not other apoptosis-related features. The data suggest that M95 mutation-related cell death involves both caspase-dependent and -independent pathways.
