Glutathione-responsive copper-disulfiram nanoparticles for enhanced tumor chemotherapy

Summary: The FDA-approved drug Disulfiram has been found to have potent antitumor therapeutic effect by generating Cu(DTC)2, but its poor tumor selectivity and lack of sufficient endogenous Cu2+ concentration within the tumor site hinder its application. A GSH-responsive coordination nanoparticles (Cu-IXZ@DSF) was established to achieve selective triggered tumor in situ Cu(DTC)2 generation for antitumor therapy, showing good biosafety and excellent antitumor effect via endoplasmic reticulum stress (ERS) and reactive oxygen species (ROS) generation pathway. This nanocarrier provides a specific-triggered antitumor Cu(DTC)2 generation for DSF-based chemotherapy with high therapeutic effect and biosafety, showing great potential of treating cancer.

MMP-responsive transformation nanomaterials with TAP antagonist to boost immune checkpoint therapy

Summary: The study developed enzyme-responsive PVA-peptide conjugates (PPCs) to improve the blocking efficiency of immune checkpoints in tumor-specific T cells. The self-assembled PPC-1 nanoparticles were able to enter tumor environment and effectively interrupt the PD-1/PD-L1 interaction. By introducing IAP antagonists, the study also increased the infiltration of T cells in tumors, improving the biodistribution and accumulation of PD-L1 antagonists and achieving higher tumor growth inhibition efficiency.

TAMs are depleted by releasing the colony-stimulating factor 1 receptor (CSF1R) inhibitor Pexidartinib and delivering anti-PD-1 antibody-conjugated platelets locally and systemically, effectively inhibiting post-surgery tumor recurrence.

A cyclodextrin-based nanoformulation achieves co-delivery of ginsenoside Rg3 and quercetin for chemo-immunotherapy in colorectal cancer

Summary: This study confirms the potential of ginsenoside Rg3 as an inducer of immunogenic cell death (ICD) against colorectal cancer (CRC) cells. The effectiveness of Rg3 in inducing ICD is significantly enhanced when combined with quercetin (QTN) that induces reactive oxygen species (ROS). A targeted nanoparticle formulation, co-encapsulating Rg3 and QTN, is developed to improve the in vivo delivery and shows enhanced tumor targeting and prolonged blood circulation in a CRC mouse model. The combination of this nanoparticle formulation with anti-PD-L1 leads to significantly longer survival in animals.
