Inhibition of NPC1L1 disrupts adaptive responses of drug-tolerant persister cells to chemotherapy

Summary: The study identified a drug-tolerant persister (DTP) state in multidrug-resistant (MDR) cancer cells as a tolerance response to combination treatment, partially preventing chemotherapy-induced oxidative stress by promoting vitamin E uptake. The NRF2 transcriptionally regulated NPC1L1 by binding to its promoter, and decreased DNA methylation was also related to this process. Triple-combination therapy with chemotherapeutic agents, verapamil, and ezetimibe demonstrated significant anti-tumor effects and prevented tumor recurrence in mice.

Summary: In this study, a nanomedicine based on the tumor microenvironment (TME) was designed and developed for effective cancer treatment. The nanocomposite with a cancer cell membrane (CM) coating can evade the immune clearance and accumulate at the tumor site. Activation of the nanocomposite in the TME triggers a series of reactions, promoting cancer cell death and enhancing photodynamic therapy (PDT) efficacy. This sequential synergistic therapy has demonstrated the ability to inhibit cancer cell proliferation effectively.
