Elucidation of kinetic and structural properties of eye lens ζ-crystallin: an in vitro and in silico approach

Md Amiruddin Hashmi et al.

Summary: This study investigated the characteristics and structure of Arabian Camelus dromedarius eye lens zeta-crystallin, analyzed the enzyme kinetics parameters and physico-chemical parameters in 9,10-phenanthrenequinone substrate and NADPH cofactor, and established proper characterization. The structure of the recombinant zeta-crystallin was validated through homology modeling and molecular dynamics simulation.

Biophysical characterization of structural and conformational changes in methylmethane sulfonate modified DNA leading to the frizzled backbone structure and strand breaks in DNA

Mohd Mustafa et al.

Summary: This study utilized various techniques to investigate the structural alterations in DNA induced by MMS, including exposure of chromophoric groups, conformational changes, fragmentation, and reduced stability. The results suggest direct strand breaks in DNA due to backbone loss, along with an exothermic interaction between MMS and DNA with moderate affinity. Molecular docking and simulations indicated interactions at both the level of nitrogenous bases and DNA backbone.

Evidence for Dual Site Binding of Nile Blue A toward DNA: Spectroscopic, Thermodynamic, and Molecular Modeling Studies

Mukti Mohammad et al.

Summary: The study found that Nile Blue binds to the DNA helix by two types of modes simultaneously, with binding driven by negative enthalpy and positive entropy changes. At low concentrations, Nile Blue binds to a DNA minor groove, while it switches to intercalation mode at higher concentrations. The thermodynamic study showed that the overall binding free energy is a combination of several negative and positive free energy changes.

Hydroxyl radical induced structural perturbations make insulin highly immunogenic and generate an auto-immune response in type 2 diabetes mellitus

Mohd Talha et al.

Summary: This study characterizes the modifications of insulin induced by hydroxyl radicals, leading to structural changes, cytotoxic aggregation, and enhanced immunogenicity. These alterations amplify autoimmune reactions in type 2 diabetes patients, eliciting a humoral response.

Characterization of human serum albumin modified by hair dye component, 4-chloro-1,2-phenylenediamine: Role in protein aggregation, redox biology and cytotoxicity

Mohd Sharib Warsi et al.

Summary: Hair dye component 4-chloro-1, 2-phenylenediamine (4-Cl-o-PDA) has a strong potential in modifying human serum albumin (HSA) structure, leading to structural damage, generation of reactive oxygen species, and increased genotoxicity and cytotoxicity. Antioxidants have shown to mitigate the damaging effects of 4-Cl-o-PDA on proteins.
