Targeted inhibition of Rev-erb-α/β limits ferroptosis to ameliorate folic acid-induced acute kidney injury

Lianxia Guo et al.

Summary: The study showed that Rev-erb-alpha and Rev-erb-beta play crucial roles in regulating AKI and ferroptosis, and targeted inhibition of these molecules can alleviate AKI. Misregulation of circadian clock components may contribute to the occurrence of ferroptosis, exacerbating the severity of AKI.

Mortality of septic shock patients is associated with impaired mitochondrial oxidative coupling efficiency in lymphocytes: a prospective cohort study

Wagner Luis Nedel et al.

Summary: Septic shock is a life-threatening condition that challenges immune cells to reprogram their mitochondrial metabolism, potentially leaving metabolic signatures that can predict disease progression and clinical outcomes. By assessing mitochondrial respiration and coupling efficiency at different time points, such as ICU admission and days after, it is possible to identify patients at risk of long-term deterioration and mortality. Adjusting for possible confounders, mitochondrial coupling efficiency was found to be independently associated with 6-month mortality in septic shock patients.

Neutrophils Fuel Effective Immune Responses through Gluconeogenesis and Glycogenesis

Pranvera Sadiku et al.

Summary: Recent research has shown that neutrophils can effectively survive and kill bacteria in injured and infected tissues by generating intracellular glycogen stores through gluconeogenesis and glycogenesis. Additionally, reduced glycogen cycling in neutrophils from patients with chronic obstructive pulmonary disease may be associated with impaired function.

Inhibition of aerobic glycolysis alleviates sepsis-induced acute kidney injury by promoting lactate/Sirtuin 3/AMPK-regulated autophagy

Chuyi Tan et al.

Summary: Inhibition of aerobic glycolysis protects against sepsis-induced AKI by promoting autophagy via the lactate/SIRT3/AMPK pathway. Sepsis or LPS enhance aerobic glycolysis, while the inhibitor 2-DG downregulates glycolysis and improves kidney injury induced by sepsis by enhancing cellular autophagy.

Summary: Sepsis is a disease characterized by a dysregulated host response to infection, with both inflammation and immune suppression occurring, and no specific treatment has been validated. The challenge lies in the complexity and heterogeneity of sepsis, requiring the selection of homogenous sepsis subgroups for testing experimental therapies. Artificial intelligence may help in identifying the most suitable subgroup for specific pathway inhibitors.
