Orthogonal fluorescent chemogenetic reporters for multicolor imaging

Alison G. Tebo et al.

Summary: Spectrally separated fluorophores enable the observation of multiple targets in living cells simultaneously for a deeper understanding of cell functions. Engineered fluorogen-activating tags based on the FAST system offer orthogonality in recognition and spectral properties, allowing efficient multicolor imaging of proteins in live cells.

Color Tuning of Fluorogens for FAST Fluorogen-Activating Protein

Ivan N. Myasnyanko et al.

Summary: By using benzylidene imidazolone core, a range of color-shifted fluorogenic ligands for FAST protein were developed without compromising binding efficiency or utility for live-cell protein labeling. This study emphasizes the potential of benzylidene imidazolone derivatives for rapidly expanding live-cell fluorogenic labeling tools.

SuperPlotsOfData-a web app for the transparent display and quantitative comparison of continuous data from different conditions

Joachim Goedhart

Summary: Plots and charts are important graphical tools for making data understandable to humans. The oversimplification of data by only plotting statistical summaries can conflict with transparent communication of results. The superplot is a proposed method to improve the communication of experimental design and results.

Production of the biocommodities butanol and acetone from methanol with fluorescent FAST-tagged proteins using metabolically engineered strains of Eubacterium limosum

Maximilian Flaiz et al.

Summary: The addition of FAST as an oxygen-independent fluorescent reporter protein expands the genetic toolbox of E. limosum. Our results show that FAST-tagged fusion proteins can be constructed without negatively impacting the stability, functionality, and productivity of the resulting enzyme. Butanol and acetone can be produced from methanol using recombinant E. limosum strains expressing genes encoding fluorescent FAST-tagged fusion proteins.
