Default mode network integrity changes contribute to cognitive deficits in subcortical vascular cognitive impairment, no dementia

Qi Qin et al.

Summary: This study investigated white matter microstructural changes in the DMN in VCIND patients and found that the integrity of DMN white matter is significantly impaired in subcortical VCIND patients, which correlates with cognitive dysfunction. This disrupted DMN connectivity could explain attention, language and executive dysfunction, suggesting that DMN white matter integrity may be a neuroimaging marker for VCIND.

Summary: Carrying the epsilon 2 allele can reduce beta-amyloid accumulation in the presence of the epsilon 4 allele. This protective outcome may be crucial for the development of potential Alzheimer's disease treatments.

In vivo staging of regional amyloid progression in healthy middle-aged to older people at risk of Alzheimer's disease

Fedor Levin et al.

Summary: The study found that amyloid accumulates progressively from associative to primary neocortex, gradually involving subcortical regions. The longitudinal PET data supported the progression of amyloid as estimated cross-sectionally. In the cognitive follow-up analysis, amyloid stage II showed a faster decline in executive function, while advanced amyloid stages (III and IV) showed a faster decline across multiple cognitive domains compared to stage 0.

Subcortical and Cortical Regions of Amyloid-β Pathology Measured by 11C-PiB PET Are Differentially Associated with Cognitive Functions and Stages of Disease in Memory Clinic Patients

Summary: In memory clinic patients, five subcortical and cortical regions with A beta pathology are differentially associated with cognitive functions and stages of disease. Education has a positive effect on cognitive tests, while APOE status has a negative effect.
