Precision Nomothetic Medicine in Depression Research: A New Depression Model, and New Endophenotype Classes and Pathway Phenotypes, and A Digital Self

Michael Maes

Summary: This paper discusses the use of machine learning in depression research and the development of precision psychiatry. Precision psychiatry aims to customize the best medical treatments based on individual's genomics, environmental, and lifestyle data. The paper outlines three steps in the development of precision medicine and highlights the challenges in developing precision psychiatry.

New Drug Targets to Prevent Death Due to Stroke: A Review Based on Results of Protein-Protein Interaction Network, Enrichment, and Annotation Analyses

Michael Maes et al.

Summary: This study analyzed established biomarkers in ischemic stroke (IS) patients to identify crucial factors associated with death, including immune and hemostasis-related proteins. The immune subnetwork of IS death was strongly linked to pathways such as TLR2/4 and TNF, while the hemostasis network was enriched in processes like platelet activation and fibrin clot formation.

Multivariate Analysis of Structural and Functional Neuroimaging Can Inform Psychiatric Differential Diagnosis

Drozdstoy Stoyanov et al.

Summary: Traditional psychiatric diagnosis has faced challenges in bridging the gap between neuro-biological and clinical assessments, leading to discrepancies under conventional statistical frameworks. To address this, a novel machine learning technique called multivariate linear method (MLM) was designed to capture convergent data from voxel-based morphometry, functional resting state and task-related neuroimaging, as well as relevant clinical measures. Results from applying MLM analysis in patients with schizophrenia compared to depression show that combining neuroimaging and clinical data can inform differential diagnosis with incremental validity.

How to construct neuroscience-informed psychiatric classification? Towards nomothetic networks psychiatry

Drozdstoy Stoyanov et al.

Summary: Psychiatry is in a state of crisis, with various competing approaches to psychiatric illness and unreliable current classifications. The new nomothetic network psychiatry approach utilizes machine learning to build data-driven causal models of mental illness.

Effective Connectivity between Major Nodes of the Limbic System, Salience and Frontoparietal Networks Differentiates Schizophrenia and Mood Disorders from Healthy Controls

Sevdalina Kandilarova et al.

Summary: This study investigated the differences in connectomes between psychiatric patients and healthy controls, revealing that dysfunction in the self-regulation of the salience network may underpin major mental disorders. Key connectome features differentiate mood disorders from schizophrenia, and can serve as potential imaging biomarkers.

Periventricular White Matter Hyperintensity in Males is Associated with Post-Stroke Depression Onset at 3 Months

Xuan-qiang Tu et al.

Summary: The study found that the baseline PVWMH in male patients was an independent risk factor for PSD at 3 months, especially for late-onset PSD. This suggests that sex differences may influence the pathogenesis of PSD.
