Correlationship of Drug-Polymer Miscibility, Molecular Relaxation and Phase Behavior of Dipyridamole Amorphous Solid Dispersions

Summary: This study investigated the correlationship among quantitative drug-polymer miscibility, molecular relaxation, and phase behavior of dipyridamole (DPD) amorphous solid dispersions (ASDs) with co-povidone (CP), hydroxypropyl methylcellulose phthalate (HPMC P), and hydroxypropyl methylcellulose acetate succinate (HPMC AS). The results demonstrated a significant influence of drug-polymer miscibility on molecular mobility and phase behavior of ASDs, providing valuable insights for designing physically stable ASDs. The findings were supported by various analytical techniques such as MDSC, X-ray micro computed tomography, and confocal laser scanning microscopy.

Real-time release testing of dissolution based on surrogate models developed by machine learning algorithms using NIR spectra, compression force and particle size distribution as input data
