Oxygen-Ozone Therapy for Reducing Pro-Inflammatory Cytokines Serum Levels in Musculoskeletal and Temporomandibular Disorders: A Comprehensive Review

Alessandro de Sire et al.

Summary: This comprehensive review investigates the modulatory effects of oxygen-ozone (O2O3) therapy on oxidative stress in patients with musculoskeletal and temporomandibular disorders (TMD). O2O3 therapy is found to have potential as an adjunct treatment in various pathological conditions characterized by chronic inflammatory processes and immune overactivation.

Is the Sex Difference a Clue to the Pathomechanism of Dry Eye Disease? Watch out for the NGF-TrkA-Piezo2 Signaling Axis and the Piezo2 Channelopathy

Balazs Sonkodi et al.

Summary: Dry eye disease is a multifactorial disorder with an unclear pathomechanism. The pathogenesis of DED may involve a Piezo2 channelopathy, leading to a quad-phasic non-contact injury mechanism on a multifactorial basis.

Effects of Combined Allogenic Adipose Stem Cells and Hyperbaric Oxygenation Treatment on Pathogenesis of Osteoarthritis in Knee Joint Induced by Monoiodoacetate

Aleksandar Juskovic et al.

Summary: This study explores the efficacy of combined treatment with allogeneic adipose-derived mesenchymal stem cells (ADMSCs) and hyperbaric oxygenation treatment (HBO) in a rat osteoarthritis (OA) model. The results show that the combination therapy significantly improves cartilage regeneration in the knee joint, reduces joint damage, and alleviates inflammation.

Amyotrophic lateral sclerosis and delayed onset muscle soreness in light of the impaired blink and stretch reflexes - watch out for Piezo2

Balazs Sonkodi et al.

Summary: ALS is a fatal neurodegenerative disease that leads to motoneuron death, while DOMS causes delayed onset muscle soreness and stiffness. Both conditions share sensory deficits and impaired proprioception, but ALS spares extraocular muscles compared to limb skeletal muscles.

Delayed Onset Muscle Soreness (DOMS): The Repeated Bout Effect and Chemotherapy-Induced Axonopathy May Help Explain the Dying-Back Mechanism in Amyotrophic Lateral Sclerosis and Other Neurodegenerative Diseases

Balazs Sonkodi

Summary: Delayed onset muscle soreness (DOMS) and amyotrophic lateral sclerosis (ALS) are proposed to be caused by acute compression axonopathy of sensory afferents in the muscle spindle, possibly due to genetic/environmental factors, damaging contractions, and neuroinflammatory processes. The dysfunctional microcircuits in these diseases can affect memory and learning, ultimately interfering with life-sustaining central pattern generators in ALS.

Knee position at the moment of bone bruise could reflect the late phase of non-contact anterior cruciate ligament injury rather than the mechanisms leading to ligament failure

Alberto Grassi et al.

Summary: The study used MRI and RSA to analyze knee position at the time of bone bruise in patients, finding that these positions deviated from the normal biomechanics of flexion-extension of the knee joint.

Repurposing cancer drugs identifies kenpaullone which ameliorates pathologic pain in preclinical models via normalization of inhibitory neurotransmission

Michele Yeo et al.

Summary: The study identifies a kinase-inhibitor, kenpaullone, as an enhancer of Kcc2 gene expression, which alleviates pain behavior in animal models and repairs neural-circuit disruption caused by elevated chloride levels. This compound was found to work by inhibiting GSK3ss and activating the Kcc2 promoter via KAISO transcription factor, offering a new approach to re-normalize disrupted inhibitory neurotransmission.

Finishing stationary cycling too early after anterior cruciate ligament reconstruction is likely to lead to higher failure

Balazs Sonkodi et al.

Summary: The article proposes a tri-phasic injury model for non-contact anterior cruciate ligament injuries, highlighting the mechano-energetic microdamage of proprioceptive sensory nerve terminals as the first phase of injury. Emphasis is placed on reinstating mitochondrial energy supply and ‘breathing capacity’ of injured proprioceptive sensory neurons in the later phase of injury.

Does Compression Sensory Axonopathy in the Proximal Tibia Contribute to Noncontact Anterior Cruciate Ligament Injury in a Causative Way?-A New Theory for the Injury Mechanism

Balazs Sonkodi et al.

Summary: The authors propose a new mechanism for non-contact anterior cruciate ligament injury, suggesting that it is caused by acute compression microinjury of entrapped peripheral proprioceptive sensory axons in the proximal tibia. They highlight the importance of primary damage during strenuous exercises and secondary damage during deceleration. Elevated prostaglandin E2, nitric oxide, and glutamate may play critical roles in the neuro-modulation, leading to mechano-energetic failure and inflammatory events.

Post Orgasmic Illness Syndrome (POIS) and Delayed Onset Muscle Soreness (DOMS): Do They Have Anything in Common?

Balazs Sonkodi et al.

Summary: Post orgasmic illness syndrome is a rare condition with unknown pathomechanism and uncertain treatment, characterized by symptoms lasting 2-7 days post ejaculation. Proposed primary injury includes acute compression proprioceptive axonopathy in the muscle spindle and mitochondrial impairment. Similarities to delayed onset muscle soreness exist, but differences arise from spermidine depletion-induced physiological responses. Longitudinally, cognitive and memory consequences are noted due to learning and memory-related primary injury.

Pharmacological Treatment for Acute Traumatic Musculoskeletal Pain in Athletes

Alessandro de Sire et al.

Summary: Research indicates that there are evidence gaps for the use of analgesics for sports injuries in training and competition settings, necessitating further high-quality research. The role of pharmacological therapy in reducing acute traumatic pain in athletes should still be addressed to minimize the timing of return to sport.

More than movement: the proprioceptive system as a new regulator of musculoskeletal biology

Bavat Bornstein et al.

Summary: The proprioceptive system is not only essential for motor control, but also plays a role in musculoskeletal development, function and pathology. Understanding the underlying molecular mechanisms is crucial for promoting both basic science and medical innovations. Recent evidence suggests that dysfunction of the proprioceptive system may contribute to various musculoskeletal pathologies.
