Epithelial-mesenchymal transition and cancer stem cells: A route to acquired cisplatin resistance through epigenetics in HNSCC

Summary: Chemoresistance is associated with tumor recurrence, metastases, and short survival. In this study, the researchers established cisplatin-resistant cell lines to investigate the role of epigenetics and biological differences in the progression of head and neck squamous cell carcinoma (HNSCC) after treatment. They found that cisplatin activates the epigenetic machinery and leads to changes in cellular shape and epithelial-mesenchymal transition (EMT). The upregulation of BMI-1 and KI-67 in the more resistant cell lines suggests increased tumor aggressiveness. The researchers also observed that HDAC1 and ZEB1 play opposite roles in the development of chemoresistance, with HDAC1 being down-regulated and ZEB1 being up-regulated. Additionally, the population of cancer stem cells (CSC) increased significantly with chemoresistance, accompanied by the down-regulation of HDAC1, HDAC2, and SIRT1, as well as the accumulation of Vimentin and ZEB1.

Phenylethyl Isothiocyanate: A Bioactive Agent for Gastrointestinal Health

Summary: The incidence of gastrointestinal pathologies, particularly cancer, has been increasing, resulting in significant morbidity, mortality, and economic burden on the healthcare system. Natural phytochemicals, such as phenylethyl isothiocyanate (PEITC) found in cruciferous vegetables like watercress, have shown therapeutic and preventive effects on these diseases. PEITC possesses antioxidant, anti-inflammatory, bactericidal, and anticarcinogenic properties. This review provides an overview of the role of PEITC in the prevention and treatment of oxidative/inflammatory-related gastrointestinal disorders and discusses the safe and recommended dosage of PEITC. Moreover, it proposes a framework for sustainable methodologies to obtain and stabilize PEITC for industrial use. Further scientific studies are needed to develop PEITC products for the nutraceutical, pharmaceutical, or food industries.

Activation of ABCC Genes by Cisplatin Depends on the CoREST Occurrence at Their Promoters in A549 and MDA-MB-231 Cell Lines

Summary: In this research, the CoREST complex is identified as a key factor that controls the expression of ABC transporters in cisplatin-treated cancer cells, preventing multidrug resistance. The CoREST complex occupancy at gene promoters suppresses the EP300-dependent increase in ABCC transcription induced by cisplatin and gene overexpression in cisplatin-resistant phenotypes. The EP300-mediated activation of ABCC10 in response to cisplatin is only possible in the presence of p53.

Nutraceuticals and functional foods, including sulforaphane, provide safer alternatives to prevent or delay cancer onset amid the prevalence of lifestyle-related disorders and environmental toxins.

This review discusses the synthesis of sulforaphane and its analogues, as well as their biological activity. The mechanisms of action for anticancer and antibacterial effects are also explored.
