Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Summary: The study suggests that high circulating tumor cell count can be a reliable biomarker for guiding the choice between chemotherapy and endocrine therapy as first-line treatment for hormone receptor-positive, ERBB2-negative metastatic breast cancer.

Prognostic relevance of the HER2 status of circulating tumor cells in metastatic breast cancer patients screened for participation in the DETECT study program

V Mueller et al.

Summary: CTC detection is a strong prognostic factor in patients with HER2-negative breast cancer. The presence of >= 1 CTC with strong HER2 staining is associated with shorter overall survival.ER and PR positive tumor patients are more likely to harbor CTCs with strong HER2 staining.

Integrative statistical analyses of multiple liquid biopsy analytes in metastatic breast cancer

Corinna Keup et al.

Summary: The study analyzed multiple LBAs in 26 HR-positive, HER2-negative MBC patients, finding that using multiple LBAs increased the prevalence of patients with actionable signals. The ELIMA.score showed a significant correlation with overall survival, indicating the additive value of using multiple LBAs. Mutual information calculation revealed that none of the LBAs is independent of the others, with CTC gDNA partially describing the other three LBAs.

A chimeric virus-based probe unambiguously detects live circulating tumor cells with high specificity and sensitivity

Xinping Fu et al.

Summary: The novel method based on a chimeric virus probe can detect circulating tumor cells with extremely high specificity and sensitivity, exclusively detecting live cells and not being affected by EpCAM expression variation. The probe, composed of a capsid from human papillomavirus and an SV40-based genome, allows for high specificity and extensive amplification inside cells for high sensitivity. Marker genes can be incorporated into the probe for increased versatility in detection.

Multimodal liquid biopsy for early monitoring and outcome prediction of chemotherapy in metastatic breast cancer

Amanda Bortolini Silveira et al.

Summary: Circulating tumor cells (CTCs) and circulating tumor DNA (ctDNA) are two distinct blood biomarkers with complementary prognostic values in metastatic breast cancer patients, a comprehensive liquid biopsy approach involving simultaneous detection of ctDNA and CTCs may offer better clinical insights for patient management.
