Encapsulating Cas9 into extracellular vesicles by protein myristoylation

Summary: CRISPR/Cas9 genome editing is a promising method for treating genetic diseases. However, encapsulating the CRISPR/Cas9 machinery for delivery is challenging. In this study, the researchers found that myristoylated proteins were preferentially encapsulated into extracellular vesicles (EVs). By fusing a myristoylated peptide onto the N-terminus of Cas9, they were able to promote the myristoylation and encapsulation of Cas9 into EVs. Additionally, introducing VSV-G increased the transfection potential. The encapsulated Cas9 accounted for 0.7% of total EV protein. The EVs coated with VSV-G encapsulating Cas9/sgRNA-eGFP showed up to 42% eGFP knock out efficiency in recipient cells, with limited off-target effects.

The CUMS-responsive lncRNA DARS-AS1 was found to be enriched in TNBC clinical tumors and cells, positively correlated with late clinical stage, and overexpression of DARS-AS1 enhanced migration and invasion of TNBC tumors. Treatment with DARS-AS1 siRNA-loaded exosomes slowed CUMS-induced TNBC cell growth and liver metastasis.
