Antibodies to a Citrullinated Porphyromonas gingivalis Epitope Are Increased in Early Rheumatoid Arthritis, and Can Be Produced by Gingival Tissue B Cells: Implications for a Bacterial Origin in RA Etiology

Natalia Sherina et al.

Summary: Based on the epidemiological link between periodontitis and rheumatoid arthritis (RA), this study investigated the production of anti-citrullinated protein antibodies (ACPA) triggered by the periodontal bacterium Porphyromonas gingivalis in the gum mucosa of early RA patients. The findings suggest that immunity to P. gingivalis derived citrullinated antigens in the inflamed gum mucosa may contribute to the presence of ACPA in RA patients through mechanisms of molecular mimicry.

Novel Functions of Integrins as Receptors of CD154: Their Role in Inflammation and Apoptosis

Ghada S. Hassan et al.

Summary: CD154, an inflammatory mediator, interacts with integrins and affects various biological processes such as thrombus formation, cell activation, and release of pro-inflammatory mediators. This article summarizes the interactions between different integrins and CD154 and their importance in pro-inflammatory and apoptotic responses.

Molecular Analysis of L-Asparaginases for Clarification of the Mechanism of Action and Optimization of Pharmacological Functions

Marina Pokrovskaya et al.

Summary: L-asparaginases are enzymes that hydrolyze L-asparagine to L-aspartic acid and ammonia. They are widely used in the treatment of lymphoblastic leukemias, but their medical use is limited by side effects and immune reactions. Gene-editing methods are used to optimize these enzymes.

Summary: This article reviews the structural and biochemical properties of l-asparaginases, focusing on bacterial-type enzymes from various organisms. Structural models of over 100 l-asparaginases have been deposited in the Protein Data Bank in the last 30 years. The elucidation of the enzymatic action mechanism and ongoing evaluation of substrate specificity are contributing to efforts to enhance the clinical properties of this important anticancer drug.

Mining the capacity of human-associated microorganisms to trigger rheumatoid arthritis-A systematic immunoinformatics analysis of T cell epitopes

Jelena Repac et al.

Summary: Autoimmune diseases affect about 5% of the worldwide population, with Rheumatoid Arthritis (RA) comprising a significant proportion and showing an increasing trend. Molecular mimicry, driven by sequence similarity between microbial and self-peptides, is recognized as the primary mechanism underlying infection-induced autoimmunity. Comprehensive immunoinformatics analysis in this study reveals a larger repertoire of candidate RA-triggering organisms and identifies Endoplasmic reticulum chaperone BiP as the most likely mimicked RA-related autoantigen.

In Rheumatoid Arthritis Patients, HLA-DRB1*04:01 and Rheumatoid Nodules Are Associated With ACPA to a Particular Fibrin Epitope

Guillaume Larid et al.

Summary: In this study of 184 ACPA-positive RA patients, it was found that anti-alpha 505-515cit antibodies were associated with HLA-DRB1*04:01, and high levels of these antibodies were associated with rheumatoid nodules. This suggests that immune complexes containing these antibodies may play a role in the development of rheumatoid nodules, offering a new therapeutic opportunity for patients with this condition.
