The Multifaced Role of STAT3 in Cancer and Its Implication for Anticancer Therapy

Manlio Tolomeo et al.

Summary: STAT3 is a complex regulator of transcription, with constitutive activation reported in many tumors but less frequent somatic mutations in cancer. Oncogenic targets of STAT3 have been identified, and inhibitors have been developed for cancer prevention and treatment. However, data suggest that STAT3 functions are multifaceted and its cellular role is determined by various signals and factors, making targeted therapies tailored to specific tumor types important.

ER Stress and Unfolded Protein Response in Leukemia: Friend, Foe, or Both?

Kelly Feral et al.

Summary: The unfolded protein response (UPR) is an adaptive signaling pathway triggered by ER stress, allowing restoration of protein homeostasis and cell survival. In cancer cells and leukemias, UPR plays a critical role in cytoprotection and resistance to chemotherapy.

Targeting MCL-1 in cancer: current status and perspectives

Haolan Wang et al.

Summary: MCL-1, an antiapoptotic protein, is frequently overexpressed in many tumor types and closely associated with tumorigenesis, poor prognosis, and drug resistance. Its central role in regulating the mitochondrial apoptotic pathway makes it an attractive target for cancer therapy. Progress has been made towards MCL-1 inhibitors and some have entered clinical trials.

The Immune Regulatory Role of Protein Kinase CK2 and Its Implications for Treatment of Cancer

Huixian Hong et al.

Summary: Protein kinase CK2 is a constitutively active serine/threonine kinase that regulates cellular signaling pathways through phosphorylating proteins, contributing to cell survival, proliferation, and inflammation. It has unique functions in immune cells and targeting CK2 activity may impact cancer treatment, particularly through T-cell modulation.

Cholangiocarcinoma

Samantha Sarcognato et al.

Summary: Liver cancer is the third leading cause of cancer-related death worldwide. Cholangiocarcinoma (CCA) is the second most common type of liver cancer, with increasing incidence and mortality. Most CCAs are diagnosed at an advanced stage with limited treatment options, and they can be classified into intrahepatic, perihilar, and distal CCA.
