KDM1A inactivation causes hereditary food-dependent Cushing syndrome

Anna Vaczlavik et al.

Summary: This study identified KDM1A gene inactivation as the cause of food-dependent Cushing syndrome (FDCS) in 90% of primary bilateral macronodular adrenal hyperplasia (PBMAH) cases. Through multiomics analysis of patient tissues, the study classified patients into different molecular groups and suggested genetic screening for ARMC5 and KDM1A for earlier diagnosis and improved management.

The reactome pathway knowledgebase 2022

Marc Gillespie et al.

Summary: The Reactome Knowledgebase, a core resource of Elixir, provides manually curated molecular details of human biological processes, serving as a digital archive and tool for discovering functional relationships. Recent work has expanded annotations of signaling processes, drugs targeting them, infections by coronaviruses, and better analysis tools for high-throughput data and placement of unstudied proteins.

Summary: Through miRNA profiling, we identified miRNA signatures in patients with familial and sporadic PMAH. These differentially expressed miRNAs may be involved in the pathogenesis of PMAH. Specific miRNAs may serve as new targets for further functional studies of PMAH.

Selection and verification of the combination of reference genes for RT-qPCR analysis in rat adrenal gland development

Xuan Xia et al.

Summary: The study selected seven reference genes and used different software for analysis to select the optimal combination of reference genes. The results showed that TBP + I3-actin is the best combination for fetal rat adrenals under physiological conditions, with sex differences in infancy and adolescence, while the reference gene combination in the PCE model was consistent with physiological conditions.

PANTHER version 16: a revised family classification, tree-based classification tool, enhancer regions and extensive API

Huaiyu Mi et al.

Summary: PANTHER is a resource for evolutionary and functional classification of protein-coding genes, with extensive improvements made in the past two years, including the refactoring of Protein Class ontology and addition of services for programmatic access. The resource provides comprehensive classification and annotation information for genes from different taxonomic groups and also allows users to add their own protein sequences for evolutionary context and fine-grained annotations.

Wnt-Signaling Regulated by Glucocorticoid-Induced miRNAs

Henriett Butz et al.

Summary: The study found that glucocorticoid treatment led to miRNA expression changes in different cell types. 19.5% of the influenced signaling pathways were common in all experiments, with the Wnt signaling pathway being the most affected. Transcriptome data and tissue-specific target prediction showed similar results, indicating significant alterations in the Wnt pathway in both the GC-producing adrenal gland and all investigated GC target tissue types.

Differential microRNA Expression in USP8-Mutated and Wild-Type Corticotroph Pituitary Tumors Reflect the Difference in Protein Ubiquitination Processes

Mateusz Bujko et al.

Summary: USP8 mutations in pituitary tumors have a direct impact on the proteome and gene expression. MicroRNA profiles in USP8-mutated and wild-type pituitary tumors were compared, revealing differences in expression. However, these microRNA expression differences do not correspond to variations in gene expression between the two types of tumors.

Molecular Genetic and Genomic Alterations in Cushing's Syndrome and Primary Aldosteronism

Crystal D. C. Kamilaris et al.

Summary: This review summarizes the genetic alterations that lead to the development of benign glucocorticoid and/or mineralocorticoid producing adrenocortical tumors and hyperplasias over the past two decades through advancements in genomics. It discusses the significance of intracellular calcium signaling and abnormal cyclic adenosine monophosphate-protein kinase A signaling in aldosterone and cortisol hypersecretion, identifying key causative defects in various genes. Additionally, it highlights the role of germline pathogenic variants in ARMC5 as a tumor suppressor in cortisol-producing primary bilateral macronodular adrenal hyperplasia.

Summary: This study identified miR-182-5p as a potential biomarker for Cushing's Disease (CD), which needs further validation in a prospective cohort. The results suggest that the presence or absence of ACTH may be as relevant for miRNA expression as hypercortisolism itself in CS.
