Cross talk between LAM cells and fibroblasts may influence alveolar epithelial cell behavior in lymphangioleiomyomatosis

Debbie Clements et al.

Summary: This study found that AT2 cells in LAM exhibit a proliferative and activated phenotype, and the accumulation of LAFs plays a role in altering the behavior of AT2 cells. Fibroblast-derived FGF7 may contribute to the cross talk between LAFs and epithelial tissue.

Summary: 5-azaC can attenuate oxidant-induced senescence, stimulate autophagic response without pro-apoptotic action, and increase necrotic cell death in cells with higher nitric oxide levels under oxidative stress conditions.

Differential Modulation of Matrix Metalloproteinases-2 and -7 in LAM/TSC Cells

Silvia Ancona et al.

Summary: The study investigated the expression, secretion, and activity of MMP-2 and MMP-7 in LAM, as well as their impact on cell migration and response to drugs such as rapamycin and anti-EGFR antibody. The findings suggest that MMP-2 and MMP-7 in LAM/TSC cells are dependent on tuberin expression, cellular adhesion, and migration, and are sensitive to rapamycin and anti-EGFR antibody during cellular migration.

Physiological hypoxia restrains the senescence-associated secretory phenotype via AMPK-mediated mTOR suppression

Thijmen van Vliet et al.

Summary: Cellular senescence is a stable proliferative arrest triggered by damaging signals, and senescent cells express lower levels of detrimental pro-inflammatory SASP factors in physiologically hypoxic environments. Exposure to low-oxygen conditions leads to AMPK activation and suppression of the mTOR-NF-kappa B signaling loop, reducing SASP expression in cells and tissues. Treatment with hypoxia-mimetic compounds can improve strength in chemotherapy-treated and aged mice by reducing detrimental paracrine effects of senescent cells.
