The mitochondrial permeability transition: Recent progress and open questions

Summary: The basis of mitochondrial permeability transition, a Ca2+-dependent permeability increase of the inner membrane, has been successfully defined, and it has been recognized as a regulator of mitochondrial ion homeostasis and a mechanism of cell death. However, there are still controversial mechanisms regarding the permeability transition that require further research for explanation.

Despite almost 50 years of research, the mechanisms of mPTP are still not definitively understood. From initially being considered an in vitro artifact to now being recognized for its physiological and pathological implications, the journey of mPTP research has been long and complex.

Effects of Tl+ on the inner membrane thiol groups, respiration, and swelling in succinate-energized rat liver mitochondria were modified by thiol reagents

Summary: The study showed that both Tl+ and thiol reagents can affect the inner membrane of mitochondria by increasing swelling and decreasing content, potential, and respiration. The most pronounced effect was observed with phenylarsine oxide, which reacts with thiol groups in the hydrophobic regions of the membrane. Effective concentrations of other reagents were approximately one to two orders of magnitude higher than phenylarsine oxide.
