Distinct epilepsy phenotypes and response to drugs in KCNA1 gain- and loss-of function variants

Francesco Miceli et al.

Summary: This study compared the functional consequences of different KCNA1 gene variants in neurological diseases, revealing that pore mutations lead to loss-of-function effects, while voltage sensor mutations cause a hyperpolarizing shift in the activation process. There is a novel correlation between in vitro phenotypes (GoF vs LoF) and clinical presentation (mild vs severe).

Lacosamide effectiveness and tolerability in patients with drug-resistant epilepsy and severe disability under polytherapy: Therapy optimization as emerging from an observational study

Marco Pozzi et al.

Summary: The study investigated the effectiveness and safety of lacosamide combined with inhibitors of fast-inactivated sodium channels or other antiepileptic drugs in treating drug refractory focal epilepsy associated with intellectual or psychiatric disability. The results showed that lacosamide could reduce the frequency of seizures and had good tolerability. The retention rate of lacosamide was higher in patients with severe intellectual disability.

Kv1.1 preserves the neural stem cell pool and facilitates neuron maturation during adult hippocampal neurogenesis

Yuan-Hung Lin King et al.

Summary: Adult hippocampal neurogenesis plays a critical role in learning and memory, and the K(v)1.1 channel has been identified as an important regulator in this process. Deletion of K(v)1.1 leads to overproliferation and depletion of adult neural stem cells, impairs maturation and integration of new cells in the hippocampus, and moderately affects hippocampus-dependent contextual fear learning and memory.

Musculoskeletal Features without Ataxia Associated with a Novel de novo Mutation in KCNA1 Impairing the Voltage Sensitivity of Kv1.1 Channel

Paola Imbrici et al.

Summary: Mutations in the KCNA1 gene are classically associated with episodic ataxia type 1, but can also lead to various symptoms including neuromyotonia. A novel de novo variant, T268K, was identified in this study, disrupting Kv1.1 function and causing neuromyotonia, muscle hypertrophy, and skeletal deformities.

Refining Genotypes and Phenotypes in KCNA2-Related Neurological Disorders

Jan H. Doering et al.

Summary: Pathogenic variants in KCNA2 gene can lead to a range of neurological disorders with diverse clinical manifestations, including early-onset developmental and epileptic encephalopathy, intellectual disability, and movement disorders. Individuals with different types of variants may exhibit distinct clinical features, with carriers of complex and mixed channel dysfunction variants more likely to show early symptoms.

KCND2 variants associated with global developmental delay differentially impair Kv4.2 channel gating

Yongqiang Zhang et al.

Summary: This study reports six individuals with early-onset global developmental delay carrying heterozygous missense variants of KCND2, resulting in impaired Kv4.2 channel gating. The results support the role of Kv4.2 channel gating impairment in early-onset monogenic global developmental delay.

Kv1.1 channels regulate early postnatal neurogenesis in mouse hippocampus via the TrkB signaling pathway

Shu-Min Chou et al.

Summary: The study elucidates the important role of the Kv1.1 potassium channel in regulating the proliferation of postnatal neural progenitor cells in the mouse hippocampus. Loss of Kv1.1 channel activity leads to depolarization of neural progenitor cells and promotes their proliferation.

Therapeutic Potential of Sodium Channel Blockers as a Targeted Therapy Approach in KCNA1-Associated Episodic Ataxia and a Comprehensive Review of the Literature

Stephan Lauxmann et al.

Summary: Variants in the KCNA1 gene causing episodic ataxia type 1 are associated with neurological symptoms such as gait incoordination and nystagmus. Sodium channel blockers like carbamazepine have shown promise in restoring altered gating properties of dysfunctional K(V)1.1 channels. Personalized therapeutic regimens tailored to disease-causing mechanisms may offer improved treatment options for patients.
