Dysregulation of the Enteric Nervous System in the Mid Colon of Complement Component 3 Knockout Mice with Constipation Phenotypes

Promotion of the inflammatory response in mid colon of complement component 3 knockout mice

Summary: The current study investigates the impact of complement component 3 (C3) deficiency on receptor downstream-mediated inflammatory response. The results show that C3 knockout (KO) mice exhibit significant enhancement in the iNOS-mediated COX-2 induction pathway, inflammasome pathway, NF-kappa B activation, and inflammatory cytokine expressions. Additionally, lower levels of E-cadherin, tight junction channels, and ion channels are observed in the mid colon of C3 KO mice, along with a slight increase in myeloperoxidase (MPO) activity for neutrophils.

Loperamide-induced Constipation Activates Inflammatory Signaling Pathways in the Mid Colon of SD Rats Via Complement C3 and its Receptors

Summary: This study aimed to investigate the regulatory role of C3 and its receptors during constipation. By treating SD rats with Lop, it was found that the expressions of C3aR and C3bR were downregulated, while the downstream signaling pathways of C3 and its receptors (including PI3K/AKT, RhoA/MLC, and MAP kinase pathways) as well as inflammatory cytokine expressions were upregulated.

Further research is needed to determine if aberrant activity of CR3 and CR4 leads to altered immune responses in individuals carrying these variants, and their role in pre-eclampsia pathogenesis.
