Transcriptional Response in a Sepsis Mouse Model Reflects Transcriptional Response in Sepsis Patients

Florian Rosier et al.

Summary: In this study, the transcriptional response of mice injected with LPS was compared to human cells stimulated with LPS in vitro and the blood cells of septic patients. The results showed a significant overlap in gene regulation and disease processes between mouse and human sepsis, supporting the hypothesis of common molecular and cellular mechanisms.

Autotaxin levels in serum and bronchoalveolar lavage fluid are associated with inflammatory and fibrotic biomarkers and the clinical outcome in patients with acute respiratory distress syndrome

Lijuan Gao et al.

Summary: This study found that serum ATX, MMP-7, and BALF IL-8 levels were significantly different between ARDS patients who survived up to 28 days and those who did not. Serum and BALF levels of ATX were positively correlated with other inflammatory and fibrotic biomarkers, and serum ATX was a better predictor of ARDS-related mortality than traditional severity scores.

Dysregulation of the Lysophosphatidylcholine/Autotaxin/Lysophosphatidic Acid Axis in Acute-on-Chronic Liver Failure Is Associated With Mortality and Systemic Inflammation by Lysophosphatidic Acid-Dependent Monocyte Activation

Francesca M. Trovato et al.

Summary: This study found that ACLF patients have reduced LPC levels, elevated LPA levels, and increased ATX concentrations correlated with cytokines IL-6 and TNF-alpha. LPA suppresses MerTK/CD163 expression and enhances monocyte proinflammatory cytokine production. This metabolic pathway could be explored as a potential therapeutic target for reprogramming monocytes in ACLF.

Inhibition of Autotaxin and Lysophosphatidic Acid Receptor 5 Attenuates Neuroinflammation in LPS-Activated BV-2 Microglia and a Mouse Endotoxemia Model

Lisha Joshi et al.

Summary: The ATX/LPA/LPA-receptor axis could be a potential pharmacological target for modulating the neuroinflammatory response triggered by systemic inflammation. Inhibition of ATX and LPA5 may protect against the harmful effects of systemic endotoxemia on the brain.

Summary: The study suggests a potential role for the ATX and LPA signaling pathway in systemic inflammation and related vascular damage in COVID-19 patients. Clinical findings demonstrate that dexamethasone treatment can reduce ATX levels, potentially providing a beneficial impact on the treatment of COVID-19 patients.

Dysregulation of the Lysophosphatidylcholine/Autotaxin/Lysophosphatidic Acid Axis in Acute-on-Chronic Liver Failure Is Associated With Mortality and Systemic Inflammation by Lysophosphatidic Acid Dependent Monocyte Activation

Summary: The study revealed that patients with ACI,F had decreased levels of LPC and increased levels of LPA in serum. Up-regulation of ATX in ACI,F promoted the production of LPA from LPC, resulting in increased proinflammatory response in monocytes.
