Simple and Sensitive Method for the Quantitative Determination of Lipid Hydroperoxides by Liquid Chromatography/Mass Spectrometry

Summary: Lipid hydroperoxides (LOOH) are important products of lipid oxidation and have adverse effects on human health. In this study, a sensitive and selective analytical method for determining LOOH was developed. The method involved chemical synthesis and chemical labeling to improve stability and ionization efficiency. Application of the method to human serum samples revealed linoleic acid hydroperoxide and oleic acid hydroperoxide as the major LOOHs. Overall, the validated LC-MS/MS methodology provides an enhanced detection and rapid separation tool for quantifying multiple LOOHs in lipid peroxidation, with potential diagnostic applications.
