Iterative model-based experimental design for spherical agglomeration processes

Kanjakha Pal et al.

Summary: Spherical agglomeration is a process intensification strategy that reduces the number of unit operations in pharmaceutical manufacturing. Efficient operation requires model-based design and control strategies, while development of a high-fidelity process model involves selecting appropriate models and accurately estimating parameters.

Comparison Study of KBH4 Spherical Agglomerates Prepared in Different Antisolvents: Mechanisms and Properties

Zhengdong Zhang et al.

Summary: By utilizing the antisolvent crystallization method with the assistance of KOH, and selecting different antisolvents, spherical agglomerates of potassium borohydride with improved flowability and anticaking ability were prepared. The size, shape, and density of the spherical agglomerates can be controlled by adjusting process parameters and choosing the appropriate antisolvent.

Distinct pathways of solid-to-solid phase transitions induced by defects: the case of DL-methionine

Genpei Shi et al.

Summary: The study investigates the effects of crystal defects on solid-to-solid phase transitions between DL-methionine polymorphs, revealing a two-step phase transition process and the influence of defect density on the transition. The research highlights the significant impact of crystal defects on solid-state phase transitions and proposes a universal two-step transition mechanism due to the ubiquitous presence of defects in crystalline materials.

Highly Polymorphous Nicotinamide and Isonicotinamide: Solution versus Melt Crystallization

Noalle Fellah et al.

Summary: The crystallization of nicotinamide (NA) and its constitutional isomer, isonicotinamide (INA), was compared in terms of polymorph generation from the melt and solution. The analysis revealed that the selectivity of polymorphs is not related to supramolecular self-association in the growth media. Kinetic analyses suggest that cross nucleation may be the reason for more polymorphs of NA crystallizing from the melt compared to INA.

Spherical Agglomerates of m-Aminobenzoic Acid: Solvent Selection, Preparation, Mechanism, and Characterization

Yiting Xiao et al.

Summary: Solvents for spherical agglomerate preparation are typically chosen based on the Lifshitz-van der Waals acid-base theory with negative adhesion free energies. However, this study found that some solvents with positive adhesion free energies can also produce spherical agglomerates. By screening solvents using m-aminobenzoic acid as a model compound, water, isopropyl alcohol, ethylene glycol, methanol, and ethanol were identified as suitable solvents for spherical crystallization. The solvents capable of forming spherical agglomerates have both hydrogen-bond-donating and hydrogen-bond-accepting capabilities. The study also investigated the effects of different factors on the formation of spherical agglomerates and elucidated the formation mechanism using in situ crystallization monitoring. Spherical agglomerates were found to exhibit better chemical stabilities and crystal form stability compared to needle-like crystals in comparative experiments.

Enabling the direct compression of metformin hydrochloride through QESD crystallization

Jerome Hansen et al.

Summary: The study showed that the QESD crystallization technique can significantly improve the flowability and reduce storage agglomeration of metformin hydrochloride, allowing for direct compression into tablets. However, the tabletability of the material needs to be evaluated under actual production speeds, with potential solutions including reducing punch speed or using a precompression step to avoid deaeration issues caused by the porous structure of metformin agglomerates. Additionally, the influence of surfactants used in QESD crystallization on the strength of tablets produced was also analyzed due to limited literature on this topic.

Pure Curcumin Spherulites from Impure Solutions via Nonclassical Crystallization

K. Vasanth Kumar et al.

Summary: Crystallization experiments on highly supercooled solutions produced highly pure and crystalline mesocrystals of curcumin, exhibiting a crystallographic hierarchy. The formation of spherulites is attributed to the high degree of supercooling and particle-mediated crystallization is proposed as an efficient method for organic compound purification.
