Decade-long leukaemia remissions with persistence of CD4(+) CAR T cells

J. Joseph Melenhorst et al.

Summary: CAR T cells redirected to target CD19 demonstrated long-lasting potential and clonal stability in two patients with chronic lymphocytic leukaemia. Highly activated CD4(+) cells emerged and dominated the CAR T cell population at later time points. These unexpected CAR T cell populations provide novel insights into anti-cancer response and long-term remission in leukaemia.

Tumor infiltrating and peripheral CD4+ILT2+ T cells are a cytotoxic subset selectively inhibited by HLA-G in clear cell renal cell carcinoma patients

Alix Jacquier et al.

Summary: Recent studies have identified HLA-G:ILT2 as a major immune checkpoint in urologic cancers, particularly in clear cell renal cell carcinoma (ccRCC). CD4+ T cells expressing ILT2 have been found in urologic cancer patients, showing a statistical association with tumor context and a positive correlation between peripheral and intra-tumoral levels. Phenotypic and functional analyses revealed that these CD4+ILT2+ T cells exhibit memory T cell and cytotoxic markers and have high cytolytic properties, functioning as proper CD4+CTLs but are selectively inhibited by HLA-G+ targets. Our findings suggest that ILT2+ T cells could be a target for novel immunotherapy in HLA-G+ tumors.

Comprehensive Profiling of an Aging Immune System Reveals Clonal GZMK+ CD8+ T Cells as Conserved Hallmark of Inflammaging

Denis A. Mogilenko et al.

Summary: This study characterized age-associated alterations in immune cells across multiple mouse organs and identified a distinct population of age-associated GZMK-expressing CD8(+) T cells. These Taa cells, highly clonal with specific features, were found to be a potential target to address age-associated dysfunctions of the immune system. In humans, proportions of the circulating GZMK(+) CDE3(+) T cell population that shares signatures with mouse Taa cells also increased during healthy aging.

A novel CD4+CTL subtype characterized by chemotaxis and inflammation is involved in the pathogenesis of Graves' orbitopathy

Yue Wang et al.

Summary: Research has revealed a novel cell type called CD4+ cytotoxic T lymphocytes (CTLs) that play crucial roles in driving Graves' orbitopathy (GO). These CTLs exhibit chemotactic and inflammatory features, show strong cytotoxic response to autoantigens, localize in orbital sites, and potentially relate to disease relapse, providing new insights into the pathogenesis of GO.

Involvement of cytotoxic Eomes-expressing CD4+ T cells in secondary progressive multiple sclerosis

Ben J. E. Raveney et al.

Summary: This study identified a subset of cytotoxic CD4(+) T cells expressing Eomes, known as Eomes(+) Th cells, that are crucially involved in the pathogenesis of secondary progressive MS (SPMS). Eomes levels could act as a biomarker to predict disease worsening in SPMS patients with over 80% accuracy. These findings have significant implications for SPMS biomarkers and potential therapeutic targets.

Origin and fine-tuning of effector CD8 T cell subpopulations in chronic infection

Kristiyan Kanev et al.

Summary: Persisting stimulation can lead CD8 T cells towards a hypofunctional state known as T cell exhaustion, which may serve to balance viral control and immunopathology. Recent research has revealed the crucial roles of the transcription factor Tox and CD4 helper T cells in shaping exhausted CD8 T cell differentiation and effector subsets. These insights could potentially be harnessed to enhance immune responses in chronic infections and cancer.

Tumor-specific cytolytic CD4 T cells mediate immunity against human cancer

Amelie Cachot et al.

Summary: CD4 T cells displaying cytotoxic phenotypes were identified in different human cancers through mining single-cell RNA-seq datasets. Ex vivo confirmation of the cytolytic tumor-specific CD4 T cells was achieved using peptide-MHCII-multimer technology. The cytotoxic activity of these cells, partially dependent on SLAMF7, was demonstrated to have delayed kinetics compared to classical cytotoxic lymphocytes, and agonistic engagement of SLAMF7 enhanced their cytotoxicity, indicating potential synergy with other cancer immunotherapies.

CD4 T-Cell Exhaustion: Does It Exist and What Are Its Roles in Cancer?

Alexandra M. Miggelbrink et al.

Summary: In chronic infections and cancer, persistent antigen stimulation can lead to T-cell exhaustion, with CD4 T-cell exhaustion being less studied but potentially clinically relevant in orchestrating immune responses and disease severity. Understanding phenotypic exhaustion in CD4 T cells and its impact on immune responses and disease severity is crucial for identifying novel therapeutic targets in cancer.

Mechanisms of Antiviral Cytotoxic CD4 T Cell Differentiation

Cory J. Knudson et al.

Summary: This study reveals the induction and differentiation mechanisms of CD4-CTL during viral infection, showing that CD4-CTL are derived from terminal differentiation of classical Th1 cells and require sustained antigen presentation. The research demonstrates that CD4-CTL upregulate protein levels of transcription factors posttranscriptionally, suggesting further posttranscriptional regulation is necessary for CD4-CTL differentiation.

Essential role of a ThPOK autoregulatory loop in the maintenance of mature CD4+ T cell identity and function

Jayati Basu et al.

Summary: The transcription factor ThPOK plays a critical role in the homeostasis and differentiation of mature helper T cells, controlling the expression of Sil(ThPOK) in CD4 T cells through an autoregulatory loop. Disruption of this loop leads to changes in chromatin accessibility and the derepression of colonic T-reg cell gene expression, promoting the differentiation of naive CD4 T cells into specific T-reg cells and CD4(+) IELs, providing protection from colitis in the gut. This autoregulatory loop represents a key mechanism for the physiological control of ThPOK expression and T cell differentiation in the gut with potential therapeutic relevance.
