Galectin-3 regulates microglial activation and promotes inflammation through TLR4/MyD88/NF-kB in experimental autoimmune uveitis

Summary: This study investigated the effect and mechanism of Galectin-3 on microglial activation and inflammation in autoimmune uveitis. The results showed that the expression of Galectin-3 was increased in microglia of uveitis retinas. Inhibition of Galectin-3 improved the clinical and pathological manifestations of uveitis and reduced the expression of proinflammatory factors. The mechanism involved down-regulation of TLR4 and MyD88 expression and inhibition of NF-kappa B p65 activation in microglia.

Summary: This study reveals the ontogenetic relationships and development of different subsets of macrophages in the central nervous system (CNS). Microglia and meningeal macrophages share a common prenatal progenitor, while perivascular macrophages originate from the perinatal meningeal macrophages after birth in an integrin-dependent manner. The presence of arterial vascular smooth muscle cells is crucial for the establishment of perivascular macrophages.

Summary: Animal models of glaucoma show that unilateral or bilateral ocular hypertensive glaucoma can lead to increased neuroinflammatory responses, with more marked effects in animals with bilateral glaucoma. Unilateral glaucoma can also activate microglia in the contralateral eye, indicating the need for caution when using the contralateral eye as a control. Neuroinflammatory protein responses identified in this study may serve as potential disease-modifiable targets.

Summary: The TAM receptor kinases are found to be essential for microglial recognition and engulfment of amyloid plaques in Alzheimer's disease, with their phagocytosis actually promoting the formation of dense-core plaques. This study also reveals that Axl and Mer play crucial roles in microglial response to amyloid plaques, and their deficiency leads to impaired detection, response, organization, and phagocytosis of amyloid-beta plaques.

Summary: The study investigated the morphological and molecular consequences of P2X7 receptor stimulation in microglial cells, both in vivo and in vitro, as well as the receptor's contribution in a model of increased intraocular pressure. Results showed that P2X7 receptor stimulation led to rapid activation of microglial cells, including process retraction and cell body enlargement, and upregulation of markers associated with M1 and M2 activation. The findings suggest that ATP release and P2X7 receptor stimulation play a role in the early microglial response to pressure changes.

Astrocytic interleukin-3 programs microglia and limits Alzheimer's disease

Summary: Communication within the glial cell ecosystem is essential for neuronal and brain health. This study demonstrates that astrocyte-sourced interleukin-3 programs microglia to ameliorate the pathology of Alzheimer's disease by enhancing their ability to clear aggregates of beta-amyloid and tau, ultimately restricting disease progression and cognitive decline. These findings highlight IL-3 as a key mediator of astrocyte-microglia cross-talk and a potential therapeutic target for Alzheimer's disease.
