Comprehensive functional annotation of susceptibility variants identifies genetic heterogeneity between lung adenocarcinoma and squamous cell carcinoma

Na Qin et al.

Summary: This study integrated genotype data of lung adenocarcinoma and squamous cell carcinoma cases to analyze susceptibility variants for non-small cell lung cancer, revealing differences between AD and SqCC genes in pathways and functions. The results shed light on the genetic heterogeneity between AD and SqCC in NSCLC.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Summary: The review discusses emerging experimental approaches for functional studies of causal variants and translational advances from GWAS findings to disease prevention and treatment. It highlights the use of genome editing technologies in GWAS functional studies with proof-of-principle examples, and discusses challenges in interrogating causal variants and potential therapeutic opportunities.

Genome-wide association meta-analysis identifies pleiotropic risk loci for aerodigestive squamous cell cancers

Corina Lesseur et al.

Summary: Squamous cell carcinomas (SqCC) of the aerodigestive tract share common etiological risk factors and genetic susceptibility. A meta-analysis of GWAS data identified novel and pleiotropic risk variants, highlighting the importance of cross-cancer analyses. Gene-based association and gene set enrichment revealed SqCC-related genes involved in DNA damage and epigenetic regulation pathways. The study suggests overlap in genetic factors influencing aerodigestive SqCC risk in European populations.

Rare deleterious germline variants and risk of lung cancer

Yanhong Liu et al.

Summary: Recent studies indicate that rare variants could have a significant impact on the etiology of lung cancers. Through comprehensive sequencing, rare deleterious variants associated with lung cancer susceptibility were identified, including both known and novel susceptibility genes. Further mechanistic studies are needed to evaluate the pathogenic effects of these specific alleles.

RAD52: Paradigm of Synthetic Lethality and New Developments

Matthew J. Rossi et al.

Summary: DNA double-strand breaks and inter-strand cross-links are the most harmful types of DNA damage that lead to genomic instability and cancer development. Rad52, a key HR protein in eukaryotes, has been found to play critical roles in backup DNA repair pathways in HR-deficient cancer cells. Recent studies have revealed RAD52's biochemical activities and its importance in multiple DNA repair processes.

A cross-tissue transcriptome-wide association study identifies novel susceptibility genes for lung cancer in Chinese populations

Meng Zhu et al.

Summary: This study conducted a cross-tissue transcriptome-wide association study using UTMOST, identifying 6 known susceptibility genes and 12 novel ones for lung cancer susceptibility. Among these, 5 novel genes were significantly associated with lung cancer risk in both cross-tissue and lung tissue models. Further analysis indicated that specific genetic variants drove the GWAS association signals and influenced the expression of certain genes in lung tissue, supporting their role in lung carcinogenesis.
