Myeloid-Cell-Specific IL-6 Signaling Promotes MicroRNA-223-Enriched Exosome Production to Attenuate NAFLD-Associated Fibrosis

Xin Hou et al.

Summary: This study investigated the role of IL-6 signaling in NAFLD by regulating antifibrotic microRNA-223 in myeloid cells. The results showed that myeloid-specific IL-6 signaling inhibits liver fibrosis through exosomal transfer of antifibrotic miR-223 into hepatocytes. These findings provide potential therapeutic targets for the treatment of NAFLD.

Heterogeneity of HSCs in a Mouse Model of NASH

Sara Brin Rosenthal et al.

Summary: This study utilized single-cell RNA sequencing to characterize the phenotypic changes of hepatic stellate cells (HSCs) in a NASH model, revealing unexpected heterogeneity of HSC phenotypes under normal and injured states. The NASH livers showed four distinct HSC clusters, including one representing the classic fibrogenic myofibroblast, while livers with NASH regression had one cluster of inactivated HSCs similar to quiescent HSCs.

A targetable LIFR-NF-κB-LCN2 axis controls liver tumorigenesis and vulnerability to ferroptosis

Fan Yao et al.

Summary: The loss of LIFR promotes liver tumorigenesis and resistance to drug-induced ferroptosis by upregulating iron-sequestering cytokine LCN2 through NF-kappa B signaling. Targeting LCN2 with a neutralizing antibody may enhance ferroptosis induction and anticancer effects in liver cancer therapy.

Mechanisms and disease consequences of nonalcoholic fatty liver disease

Rohit Loomba et al.

Summary: NAFLD and NASH are leading chronic liver diseases worldwide, causing progressive liver injury that can lead to cirrhosis and hepatocellular carcinoma. These diseases are associated with hepatic glucose and lipid metabolism, and are influenced by genetic, epigenetic, and environmental factors.

Nonalcohol fatty liver disease: balancing supply and utilization of triglycerides

Leinys S. Santos-Baez et al.

Summary: Nonalcoholic fatty liver disease (NAFLD) is a liver condition characterized by abnormal accumulation of fats, with its incidence increasing significantly due to factors like obesity and diabetes. Recent research highlights the key role of insulin resistance, obesity, and diabetes in the development of metabolic-associated fatty liver disease (MAFLD).

Liver-fibrosis-activated transcriptional networks govern hepatocyte reprogramming and intra-hepatic communication

Anne Loft et al.

Summary: Liver fibrosis is a strong predictor of long-term mortality in individuals with metabolic-associated fatty liver disease. The study shows comprehensive alterations in hepatocyte genomic and transcriptional settings during NASH progression, leading to a loss of hepatocyte identity. The reprogramming of hepatocytes is under tight cooperative control of a network of fibrosis-activated transcription factors, promoting hepatocyte dysfunction and directing intra-hepatic crosstalk necessary for NASH and fibrosis progression.
