Synchrotron-Based Nano-X-Ray Absorption Near-Edge Structure Revealing Intracellular Heterogeneity of Iron Species in Magnetotactic Bacteria

Daniel M. Chevrier et al.

Summary: This study reports the use of scanning X-ray fluorescence microscopy (SXFM) and nanoscale X-ray absorption near-edge structure (nano-XANES) mapping to identify intracellular iron species, providing insight into the role of iron storage in magnetite biomineralization by MTB at the single-cell level.

Alzheimer's disease brain-derived extracellular vesicles spread tau pathology in interneurons

Zhi Ruan et al.

Summary: This study comprehensively characterized the physicochemical structure and pathogenic function of extracellular vesicles derived from human brains in Alzheimer's disease, prodromal Alzheimer's disease, and non-demented control cases. It was found that extracellular vesicles from Alzheimer's disease cases had higher levels of tau oligomers and exhibited more efficient propagation and misfolding of tau, suggesting a novel mechanism for tau spread in the hippocampus.

Loss of ferroportin induces memory impairment by promoting ferroptosis in Alzheimer's disease

Ferroptosis and NRF2: an emerging battlefield in the neurodegeneration of Alzheimer's disease

Darius J. R. Lane et al.

Summary: Ferroptosis is a cell death mechanism dependent on iron and lipid peroxidation, with potential implications in various neurodegenerative diseases. The main cellular protector against ferroptosis is glutathione peroxidase 4 (GPX4). The NRF2 signaling pathway plays a key role in regulating ferroptosis.

Zinc Status Alters Alzheimer's Disease Progression through NLRP3-Dependent Inflammation

Jack Rivers-Auty et al.

Summary: Alzheimer's disease is a devastating neurodegenerative disease with increasing prevalence, and inflammatory lifestyle factors may increase the risk. Zinc deficiency, the most prevalent malnutrition globally, could be a risk factor for Alzheimer's disease, while zinc supplementation may reduce the risk of cognitive decline.

Systematic Review: Quantitative Susceptibility Mapping (QSM) of Brain Iron Profile in Neurodegenerative Diseases

Parsa Ravanfar et al.

Summary: Iron has been increasingly implicated in the pathology of neurodegenerative diseases. The development of QSM technique allows for more comprehensive investigation of iron distribution in the brain. Existing studies have shown increased magnetic susceptibility in brain regions associated with different neurodegenerative disorders, indicating potential for QSM as a promising tool in investigating neurodegeneration.

Iron loading is a prominent feature of activated microglia in Alzheimer's disease patients

Boyd Kenkhuis et al.

Summary: Brain iron accumulation accelerates disease progression in Alzheimer patients with amyloid-beta. Activated microglia with increased iron accumulation influence the functional phenotype of cells, especially in conjunction with amyloid-beta. Spatial analysis showed that iron-accumulating microglia predominantly infiltrated A beta-plaques in the brain.

Synchrotron X-ray Fluorescence and FTIR Signatures for Amyloid Fibrillary and Nonfibrillary Plaques

Elena Alvarez-Marimon et al.

Summary: Amyloid plaques in the cortex of patients with advanced Alzheimer's disease have been characterized into two types: fibrillary plaques with iron accumulation mainly in Fe2+ form, and nonfibrillary plaques with a mixture of beta-sheet and unordered structures where Fe3+ predominates over Fe2+. This distinction in plaque types and metal composition could play a key role in understanding the progression of Alzheimer's disease.

Exploring valence states of abnormal mineral deposits in biological tissues using correlative microscopy and spectroscopy techniques: A case study on ferritin and iron deposits from Alzheimer's disease patients

Yitian Zeng et al.

Summary: Abnormal accumulation of inorganic trace elements in the human brain, such as iron, zinc, and aluminum, is pathologically linked to neurodegenerative diseases like Alzheimer's. Specifically, the presence of ferrous ions in brain tissues of AD patients has been associated with oxidative damage. Using physical science methods like X-ray energy dispersive spectroscopy and electron energy loss spectroscopy can provide unique insights into the chemical composition of these deposits.

Ferroptosis as a mechanism of neurodegeneration in Alzheimer's disease

Md. Jakaria et al.

Summary: Alzheimer's disease, the most prevalent form of dementia, has a complex pathophysiology that is not fully understood. While beta-amyloid plaque and neurofibrillary tangles define the disease's pathology, the mechanism of neurodegeneration remains uncertain. Ferroptosis, an iron-mediated programmed cell death mechanism, has been observed in clinical AD samples, suggesting its implication in the pathogenesis of AD.

The acute phase protein lactoferrin is a key feature of Alzheimer's disease and predictor of A beta burden through induction of APP amyloidogenic processing

Andrew Tsatsanis et al.

Summary: Amyloidogenic processing of the amyloid precursor protein leads to the formation of amyloid-beta peptide plaques in Alzheimer's disease. Early cortical changes in AD also involve neuroinflammation and elevated iron levels. The activation of the innate immune system in the brain is a neuroprotective response to infection, but persistent neuroinflammation is linked to AD neuropathology through unknown mechanisms.

Summary: Aducanumab recently underwent two large phase III clinical trials that were stopped prematurely by the sponsor due to divergent outcomes. One trial showed potential benefits from high doses of Aducanumab, while the other did not show any benefits.
