Discovery and Design of Family VIII Carboxylesterases as Highly Efficient Acyltransferases

Henrik Mueller et al.

Summary: EstCE1, identified as an efficient acyltransferase catalyzing amidation and carbamoylation in water, provides a solution to the current challenges faced by promiscuous enzymes in application. The structure-function analysis of EstCE1 reveals a three-amino acid motif essential for its promiscuous acyltransferase activity. Introducing this motif into an esterase without acetyltransferase activity can transform it into an acyltransferase.

Crystal structures of a novel family IV esterase in free and substrate-bound form

Astrid Hoeppner et al.

Summary: Bacterial lipolytic enzymes of family IV, homologs of mammalian hormone-sensitive lipases, exhibit broad substrate specificity and remarkable ability for various types of hydrolysis. Crystal structure analysis of a novel family IV esterase reveals insights into its substrate promiscuity and the important role of the cap domain in enzyme-substrate interaction. Structural data of PtEst1 are available in the worldwide protein data bank under accession codes 6Z68 and 6Z69.

T-2 Toxin-The Most Toxic Trichothecene Mycotoxin: Metabolism, Toxicity, and Decontamination Strategies

Edyta Janik et al.

Summary: T-2 toxin, the most toxic fungal secondary metabolite produced by different Fusarium species, is commonly found in contaminated cereal-based food and feed made from wheat, barley, rye, oats, and maize. Various decontamination techniques have been developed to mitigate the concentration of T-2 toxin in agricultural commodities, but complete degradation of this toxin is believed to be impossible.

Promiscuous Esterases Counterintuitively Are Less Flexible than Specific Ones

Christina Nutschel et al.

Summary: Understanding the molecular origin of observed promiscuity range in enzymes is crucial, with the study revealing that promiscuous esterases are less flexible, more thermostable, and have increased specific activity compared to specific ones. This research provides a new starting point for exploration in biotechnology and synthetic chemistry by screening esterase sequence space through rigidity analyses.

Evolution of β-lactamases and enzyme promiscuity

Christopher Frohlich et al.

Summary: Beta-lactamases are a common resistance mechanism against beta-lactam antibiotics and have become important models in enzymology and evolutionary biochemistry. They have evolved multiple times from distinct evolutionary origins, exhibiting diverse structural features and enzymatic mechanisms. Understanding their evolution and diversity can aid various disciplines.
