Trifluoroacetic Acid Promoted Ring Contraction in 2,3-Di-O-silylated O-Galactopyranosides and Hemiacetals

Summary: This study demonstrates, for the first time, the pyranose ring contraction of 2,3-di-O-silylated O-galactopyranosides in the presence of anhydrous trifluoroacetic acid, resulting in the formation of corresponding anomeric triols in furanose form. TBDPS protective groups were found to be more stable than TIPS groups under conditions of TFA-promoted pyranose ring contraction, presenting an advantage in the synthesis of selectively protected monosacharide building blocks.

Mechanistic Study of Silyl-Assist Effect on 1,2-cis-α-Glucosylation

The silyl-assist effect was mainly due to electronic factors, as shown by chemical experiments and density functional theory (DFT) calculations.

Rational design of glycosylation reactions, consideration of counterions and concentration, and collaboration among scientists from different fields are essential to address the sophisticated challenges and advance the development of stereospecific glycosylation reactions.

The Influence of Anomeric Configuration and Aglycone Structure on the Outcome of Acid-Promoted Ring Contraction in 2,3-Di-O-Silylated S-Galactopyranosides

Summary: The acid-promoted rearrangement of a series of 1-thio-alpha/beta-D-galactopyranosides with bulky triisopropylsilyl (TIPS) or tert-butyldiphenylsilyl (TBDPS) groups at O-2 and O-3, and various substituents at the S atom in anomeric position, has been studied. The reaction rates of pyranose ring contraction in S-galactopyranosides are strongly influenced by anomeric configuration and aglycone structure. This study demonstrates that the rearrangement behavior is dependent on the anomeric configuration and aglycone structure of the galactopyranosides.
