Synthesis and evaluation of a collection of purine-like C-nucleosides as antikinetoplastid agents

Jakob Bouton et al.

Summary: The kinetoplastid parasites Trypanosoma brucei, Trypanosoma cruzi and Leishmania spp. cause neglected tropical diseases, and research has focused on finding new antikinetoplastid agents. A collection of C-nucleosides with potential activity against these parasites was synthesized, and the nature of the nucleobase surrogate was found to greatly influence the compounds' selectivity profile.

6-Methyl-7-deazapurine nucleoside analogues as broad-spectrum antikinetoplastid agents

Cai Lin et al.

Summary: This study assessed the in vitro activity of modified purine nucleoside analogues against Kinetoplastid parasites, leading to the identification of analogues with activity against multiple species. These selected analogues also demonstrated sufficient stability in liver microsomes for further in vivo evaluation.

6-Methyl-7-Aryl-7-Deazapurine Nucleosides as Anti-Trypanosoma cruzi Agents: Structure-Activity Relationship and in vivo Efficacy

Cai Lin et al.

Summary: The study shows that by modifying the pyrimidine part, the 7-aryl-7-deazapurine nucleoside can retain inhibitory activity against T. cruzi amastigote growth while improving selectivity towards mammalian cells. The 7-(4-chlorophenyl) analogue exhibited promising antitrypanosomal activity in a mouse model.

Synthesis and evaluation of 3′-fluorinated 7-deazapurine nucleosides as antikinetoplastid agents

Jakob Bouton et al.

Summary: Kinetoplastid parasites rely on purine salvage for their purine demand. Evaluating the effect of introducing a 3'-fluoro substituent in 7-deazapurine nucleosides, several analogs with potent anti-parasitic activity were discovered, showing that a fluorine atom in the 3'-position is a promising modification for the discovery of anti-parasitic nucleosides.

Revisiting Pyrazolo[3,4-d]pyrimidine Nucleosides as Anti-Trypanosoma cruzi and Antileishmanial Agents

Jakob Bouton et al.

Summary: Chagas disease and visceral leishmaniasis are neglected tropical diseases with insufficient current treatments, requiring the development of new drugs. This study reports the design and synthesis of pyrazolo[3,4-d]pyrimidine nucleosides as potential antiparasitic agents, with one compound showing activity in an acute Chagas disease mouse model.
