Characterization and modulation of surface charges to enhance extracellular vesicle isolation in plasma

Hyun-Kyung Woo et al.

Summary: Extracellular vesicles (EVs) have the potential for disease diagnosis, but are often contaminated by low density lipoproteins (LDLs) when isolated from blood. By studying the difference in electric charge between EVs and LDLs, we developed a method using ion-exchange chromatography to separate EVs from LDLs. This finding improved the analytical signals in EV molecular assays.

Supermeres are functional extracellular nanoparticles replete with disease biomarkers and therapeutic targets

Qin Zhang et al.

Summary: Supermeres are a distinct type of extracellular nanoparticles that exhibit unique biological and functional properties, with a different protein and RNA composition from small extracellular vesicles and exomeres. They are enriched with cargo involved in multiple cancers, Alzheimer's disease, and cardiovascular disease, and have specific functions in vivo, suggesting their potential as circulating biomarkers and therapeutic targets.

Mitovesicles are a novel population of extracellular vesicles of mitochondrial origin altered in Down syndrome

Pasquale D'Acunzo et al.

Summary: The study identified a previously unknown population of double-membraned EVs called mitovesicles, which contain specific mitochondrial constituents and undergo changes during pathophysiological processes involving mitochondrial dysfunction. The development of a method for the selective isolation of mitovesicles opens up new possibilities for characterizing biological processes connecting EV biology and mitochondria dynamics in vivo, as well as for innovative therapeutic and diagnostic strategies.

A multiparametric extraction method for Vn96-isolated plasma extracellular vesicles and cell-free DNA that enables multi-omic profiling

Jeremy W. Roy et al.

Summary: Extracellular vesicles (EVs) are rich material for biomarker analysis, and an optimized method using Vn96 peptide has been developed for efficient isolation of EVs and cfDNA from plasma samples, allowing for downstream analysis of DNA, RNA, and protein. This multiparametric extraction protocol enhances the sensitivity and specificity of liquid biopsy tests utilizing EVs and cfDNA biomarkers for disease detection and monitoring.

Formation of a protein corona on the surface of extracellular vesicles in blood plasma

Eszter A. Toth et al.

Summary: This study found that a protein corona is formed around EVs in blood plasma, with shared EV corona proteins identified. These results help explain the commonly reported plasma protein 'contamination' of EV preparations and offer a new perspective for EV research.
