S-adenosylmethionine-dependent methyltransferase inhibitor DZNep blocks transcription and translation of SARS-CoV-2 genome with a low tendency to select for drug-resistant viral variants

Summary: We report the in vitro antiviral activity of DZNep against SARS-CoV-2, as well as its protective efficacy against infectious bronchitis virus. We found that DZNep inhibits the recruitment of hnRNPA1, which is involved in m6A modifications of SARS-CoV-2 RNA, leading to suppressed viral genome synthesis. Furthermore, DZNep also affects the synthesis of viral proteins by disrupting the interaction between viral RNA and eIF4E.
