Small-molecule drug repurposing to target DNA damage repair and response pathways

Jacqueline A. Brinkman et al.

Summary: This article discusses the role of genotoxic therapy in cancer treatment and explores the potential of repurposing existing drugs and compounds as chemo or radiosensitizers. By studying cellular mechanisms as a framework, identifying which drugs and natural products have the most potential for repurposing is highlighted. The crucial next step is to conduct clinical trials to translate laboratory discoveries into benefiting patients.

Discovery of a novel 53BP1 inhibitor through AlphaScreen-based high-throughput screening

Yanli Sun et al.

Summary: Tumor suppressor p53-binding protein 1 (53BP1) plays a crucial role in DNA Damage Repair pathways and its dysregulation is associated with various diseases including cancer. Recent studies have identified a novel and effective 53BP1-TTD inhibitor DP308, disrupting the binding between 53BP1 and H4K20me2 peptide, showing promise as a potential tool for further optimization of biological functions and gene editing therapies.

A Cas9-transcription factor fusion protein enhances homology-directed repair efficiency

Guoling Li et al.

Summary: This study presents a strategy to enhance HDR efficiency in animal cells by spatial and temporal colocalization of the donor and Cas9 through coupling the CRISPR system with the transcription factor THAP11. By appending THAP11-specific DNA binding motifs to the double-stranded donor DNA, the HDR efficiency can be significantly improved. The THAP11-fused CRISPR system achieved a more than twofold increase in HDR-mediated knock-in efficiency, and up to 6-fold increase can be achieved through combinational use with the small-molecule HDR enhancer valnemulin.

Small-molecule inhibitors of histone deacetylase improve CRISPR-based adenine base editing

Ha Rim Shin et al.

Summary: CRISPR-based base editors are widely used for nucleotide substitutions without causing DNA breaks. Efforts are being made to improve the efficiency of both cytosine and adenine base editors. A study has identified histone deacetylase inhibitors, particularly romidepsin, as a potential option to enhance base editing efficiency.

Redefining the Histone Deacetylase Inhibitor Pharmacophore: High Potency with No Zinc Cofactor Interaction

Douglas C. Beshore et al.

Summary: A novel series of histone deacetylase (HDAC) inhibitors lacking a zinc-binding moiety has been developed, showing selectivity for certain HDAC isozymes and no interaction with the catalytic zinc. These compounds are promising for further optimization due to their nonmutagenic and off-target profiles, and exhibit efficacy comparable to marketed HDAC inhibitors belinostat and vorinostat. These results suggest a need to revise the long-standing pharmacophore model of HDAC inhibitors and present a distinct class of HDAC inhibitors.

Approaches to Enhance Precise CRISPR/Cas9-Mediated Genome Editing

Christopher E. Denes et al.

Summary: Genome modification holds great potential for disease prevention or treatment, with CRISPR/Cas9 techniques showing promise in altering disease-relevant genes. Competition among DNA repair pathways can lead to undesirable editing outcomes, but small molecule modulators and engineered CRISPR/Cas proteins have been shown to enhance precision editing efficacy.

Structural and mechanistic insights into the Artemis endonuclease and strategies for its inhibition

Yuliana Yosaatmadja et al.

Summary: Artemis is an endonuclease that plays a key role in the development of B- and T-lymphocytes and in the repair of dsDNA breaks by non-homologous end-joining. Artemis has predominantly endonuclease activity, which is different from its closely related SNM1A and SNM1B nucleases that have predominantly exonuclease activity.

Efficient biallelic knock-in in mouse embryonic stem cells by in vivo-linearization of donor and transient inhibition of DNA polymerase θ/DNA-PK

Daisuke Arai et al.

Summary: In this study, an optimized HDR-mediated knock-in method for mouse embryonic stem cells was developed, which significantly improved efficiency and minimized random integrations caused by NHEJ/MMEJ. The BiPoD method achieved efficient plasmid donor insertion and increased biallelic knock-in efficiency, providing a rapid and easy way to establish biallelic knock-in mESC lines.
