Structure and transport mechanism of the human cholesterol transporter ABCG1

Summary: This study reveals the molecular mechanism of the ABCG1-driven cholesterol transport cycle. By analyzing the structure of cholesterol-bound ABCG1, it was found that a cluster of hydrophobic residues and two sphingomyelins constitute a well-defined cholesterol-binding cavity. The exit of this cavity is closed by conserved phenylalanine residues, providing a hydrophobic path for cholesterol release depending on the acceptor concentration.

Central composite optimization of ocular mucoadhesive cubosomes for enhanced bioavailability and controlled delivery of voriconazole

Summary: This study aimed to optimize cubosomal formulations loaded with voriconazole to enhance ocular bioavailability. The chitosan-coated cubosomal formulation showed high mucoadhesive properties and increased drug concentration in vitreous humor compared to voriconazole suspension. The optimized cubosomal formulation can be considered as an efficient ocular nanocarrier for voriconazole.

Development and Optimization of Terpene-Enriched Vesicles (Terpesomes) for Effective Ocular Delivery of Fenticonazole Nitrate: In vitro Characterization and in vivo Assessment

Summary: The study aimed to load fenticonazole nitrate into terpene-enriched phospholipid vesicles as a potential delivery system for ocular fungal infection. The vesicles were prepared using thin film hydration method and optimized through a full factorial design, resulting in efficient drug delivery and enhanced adhesion to eye surfaces. In vivo studies in rabbits showed higher ocular retention and safety of the prepared terpesomes for effective ocular delivery of fenticonazole nitrate.

Implementing Spanlastics for Improving the Ocular Delivery of Clotrimazole: In vitro Characterization, Ex vivo Permeability, Microbiological Assessment and In vivo Safety Study

Summary: This study aimed to encapsulate the antifungal drug clotrimazole (CLT) into spanlastics (SPs) to provide controlled ocular delivery. The optimal formulation S1 showed high encapsulation efficiency, elastic vesicles, sustained release profile, and superior inhibition of Candida albicans development compared to CLT suspension. In vivo histopathological examination confirmed the safety of S1 for ophthalmic application in rabbits, demonstrating the efficacy of SPs for ocular delivery of CLT.

Cubogel as potential platform for glaucoma management

Summary: This study investigated the potential of cubogel as an ocular dosage form to enhance the corneal permeability of Dorzolamide Hydrochloride. DZ-loaded cubosomal dispersions were prepared and characterized to determine the optimal formulation. The optimized cubogel showed promising results in terms of safety, stability, corneal delivery capability, and higher bioavailability compared to a market product.
