Treating Arterial Ageing in Patients with Diabetes: From Mechanisms to Effective Drugs

Summary: Diabetes mellitus is a major healthcare problem characterized by hyperglycemia and chronic complications. Arterial ageing in patients with diabetes is associated with endothelial dysfunction, arterial stiffness, and impaired expression of genetic longevity pathways. There are currently no specific drugs available for preventing/treating arterial ageing, but understanding the mechanisms can help guide therapy in diabetic patients.

Summary: Despite successful therapeutic strategies in the prevention and treatment of arteriosclerosis, cardiovascular complications continue to be a major clinical and societal issue worldwide. Upregulation of the osteogenic transcription factor Runx2 in the cardiovascular system plays a crucial role in driving adverse cellular events that contribute to cardiovascular pathology. This review highlights the importance of posttranslational modifications in regulating Runx2 stability and activity, as well as the impact of Runx2-mediated osteogenic transdifferentiation of vascular smooth muscle cells on vascular inflammation and the pathogenesis of arteriosclerosis.
