Association Between Posttraumatic Stress Disorder and Epigenetic Age Acceleration in a Sample of Twins

Summary: This study of 296 male twins found that twins with current PTSD showed significantly advanced DNA methylation age acceleration compared to twins without PTSD, indicating an association between PTSD and biological aging processes. Twins with current PTSD were epigenetically older than their twin brothers without PTSD across almost all measures of DNA methylation age acceleration, with estimated differences ranging between 1.6 and 2.7 biological age year-equivalents.

Epigenetic Age Acceleration and Cognitive Decline:A Twin Study

Summary: While no association was found between DNAm age acceleration and cognitive function scores at baseline, a longitudinal analysis showed that for each additional year of age acceleration using Horvath's method, there was a 3% decline in composite executive function scores and a 2.5% decline in memory function scores within twin pairs. These results suggest that middle-aged men with older DNAm age compared to their twin brothers experienced a faster rate of cognitive decline over the subsequent 11.5 years, highlighting the role of epigenetic modifications in cognitive aging.

Summary: The study found that individuals with Major Depressive Disorder (MDD) exhibit significantly accelerated cellular aging compared to healthy controls, with an excess of 2 years in GrimAge relative to their chronological age. These findings support previous suggestions of accelerated cellular aging in MDD and are the first to demonstrate this using an epigenetic metric predictive of premature mortality.
