Production and Characterization of Bio-AuNPs to Induce Synergistic Effect Against Multidrug Resistant Bacterial Biofilm

Herein we demonstrate gold nanoparticles were synthesized by conventional bioreduction approach using Clitoria ternatea L. methanolic leaf extract. Bioreduction of chloroauric acid (HAuCl4) respectively, with the C. ternatea extract, this mixer incubated at 24 h and the mixer were characterized by UV-Vis spectroscopy, FTIR, XRD, TEM, EDS results were confirmed the presence of gold ion. Further, the synthesized bio-AuNPs were found to show highest antioxidant activity when compared with standard (vitamin C) using 2, 2'-azino-bi's (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS(+)). Also, we evaluated the antimicrobial activity of bio-AuNPs against multidrug resistant Gram-negative bacteria and Gram-positive bacteria with maximum concentration (100 A mu L) was showing strong inhibitory action. In addition, the bio-AuNPs showed potential QSI activity with different concentration (25-100 A mu g/ml) against the AHL-mediated violacein production in C. violaceum (18-88 %) and biofilm formation (22-94 %), LasA protease (14-82 %), LasB elastase (15-81 %), pyocyanin (19-86 %) productions in P. aeruginosa and again it was confirmed by microscopy results and extracellular protein were also studied by SDS-PAGE method. The synthesized bio-AuNPs showed less hemolysis activity 3.75 %, compared with American Society for Testing and Materials range < 5 %, it's recommended that the bio-AuNPs were clearly much hemocompatible for drug delivery applications.
