The value of single-molecule real-time technology in the diagnosis of rare thalassemia variants and analysis of phenotype-genotype correlation

Summary: Comparing single-molecule real-time technology (SMRT) with conventional genetic diagnostic technology for rare thalassemia mutations, a study was conducted on 434 cases using both methods. SMRT was able to identify a higher number of variants compared to conventional technology, with a 9.91% increase in positive detection rate. The combination of both methods detected 485 positive alleles among 49 types of variant, and SMRT was able to identify some variants not detected by conventional methods, highlighting its significance in clinical thalassemia gene screening.

Summary: Thalassemias are the most common genetic disorders worldwide, characterized by the deficient synthesis of one or more hemoglobin (Hb) chain(s). They result from many different genetic and molecular defects and can manifest as severe or clinically silent hematologic phenotypes. Thalassemias alpha and beta are particularly prevalent in Mediterranean and surrounding regions. Traditional treatments such as RBC transfusions, iron chelation therapy, splenectomy, and hematopoietic stem cell transplantation are challenged by the rapidly evolving fields of gene therapy and gene editing strategies.
