A series of simple curcumin-derived colorimetric and fluorescent probes for ratiometric-pH sensing and cell imaging

Summary: In this study, a simple fluorescent dye called BFCUR was developed for pH sensing. By deprotonating the phenolic hydroxyl groups, BFCUR exhibited significant red-shift and enhanced fluorescence emission, allowing for ratiometric pH detection and visual pH detection using test strips. BFCUR also showed low cytotoxicity and was successfully applied for intracellular pH detection. The findings highlight the great potential of CUR-derived BFCUR as colorimetric and fluorescent probes for ratiometric pH sensing and cell imaging.

The development of POXs bearing a 4-ethynylphenyl group on the phosphorus atom allows for functionalisation with azide-tagged biomolecules via late-stage Cu-catalysed azide/alkyne cycloaddition (CuAAC), enabling target-selective labelling. A sophisticated NIR probe has been designed to have a bell-shaped pH-response and assess endosomal maturation, demonstrating the practical utility of functionalised POXs.
