DNA-Templated Bioorthogonal Reactions via Catalytic Hairpin Assembly for Precise RNA Imaging in Live Cells

Summary: In this study, we reported the design of a fluorogenic sensor that combines a bioorthogonal reaction with a DNA cascade circuit for precise RNA imaging in live cells. By triggering catalytic hairpin assembly upon target mRNA, the chemical reaction partners are brought in close proximity, resulting in high effective concentrations and efficient bioorthogonal reactions. The fluorogenic sensor demonstrated high signal-to-noise ratio and low background signals, enabling sensitive detection of target mRNA and accurate imaging in live cells.

Catalytic Activation of Bioorthogonal Chemistry with Light (CABL) Enables Rapid, Spatiotemporally Controlled Labeling and No-Wash, Subcellular 3D-Patterning in Live Cells Using Long Wavelength Light

Fluorogenic Cyclopropenones for Multicomponent, Real-TimeImaging

Selective chemical tracking of Dnmt1 catalytic activity in live cells

Summary: Enzymatic methylation of cytosine to 5-methylcytosine in DNA is a fundamental epigenetic mechanism involved in mammalian development and disease. In this study, the researchers engineered the Dnmt1 methyltransferase to catalyze the transfer of a synthetic cofactor analog onto DNA. By editing the Dnmt1 locus in mouse embryonic stem cells and using electroporation to introduce the cofactor, they were able to selectively tag Dnmt1-specific genomic targets. This new approach, called Dnmt-TOP-seq, allows for high-resolution tracking of Dnmt1 catalysis in mammalian cells and opens the door for selective studies of other methylation pathways in eukaryotic systems.

A General Strategy to Design Highly Fluorogenic Far-Red and Near-Infrared Tetrazine Bioorthogonal Probes

Summary: A novel strategy has been developed to create highly fluorescent Huaxi-Fluor probes that exhibit significantly increased fluorescence upon forming the bioorthogonal reaction product, pyridazine. These probes have large Stokes shifts and high quantum yields, with emission wavelengths fine-tunable from 556 to 728 nm by manipulating the fluorophore skeleton. Highly photo-stable and biocompatible, these probes are suitable for live cell organelle visualization without washing steps and imaging tumors in live small animals to depths of 500 μm by two-photon excitation.

Click and Bio-Orthogonal Reactions with Mesoionic Compounds

Oxime as a general photocage for the design of visible light photo-activatable fluorophores

The oxime-caged fluorophores can be oxidized to their carbonyl derivatives upon light irradiation, restoring strong fluorophore fluorescence.
