The formation of highly oxidizing high-valent intermediates presents design challenges that have not been tackled before, emphasizing the need for purposeful design and limitation of deleterious oxidation.

Biomolecular simulation identified potential interactions and binding sites near the heme cofactor, providing insight into the catalytic workings of C45.

De Novo Design of a Self-Assembled Artificial Copper Peptide that Activates and Reduces Peroxide

Summary: Copper-containing metalloenzymes are a major class of proteins in nature catalyzing various reactions, and in this study, a de novo artificial copper peptide (ArCuP) was designed to mimic the structural and functional characteristics of these enzymes within a trimeric self-assembled polypeptide scaffold. The designed ArCuP showed peroxidase activity and demonstrated a pH-dependent reactivity with optimal conditions at pH 7.5, as revealed by combined spectroscopic and electrochemical data.
