A Novel Mouse Model of Nonalcoholic Steatohepatitis Suggests that Liver Fibrosis Initiates around Lipid-Laden Macrophages

Mayuko Ichimura-Shimizu et al.

Summary: This study utilized a HFCC diet to induce a NASH-related fibrosis model in A/J mice, showing aggravated inflammation and fibrosis in the liver tissue compared to C57BL/6J mice. The presence of lipid-laden macrophage aggregation and cholesterol crystals adjacent to fibrotic lesions provided histological evidence of fibrosis initiation. Mass spectrometry analysis identified specific molecules in the aggregated macrophages, shedding light on the pathophysiology of NASH-related fibrosis.

Liver macrophages and inflammation in physiology and physiopathology of non-alcoholic fatty liver disease

Ronan Thibaut et al.

Summary: NAFLD is the hepatic manifestation of metabolic syndrome, commonly associated with type 2 diabetes. Liver macrophages play crucial roles in maintaining liver homeostasis, promoting inflammation, and mediating fibrosis during the progression of NAFLD to NASH.

Summary: This study demonstrated that the mechanisms underlying the metabolic and genetic components of NAFLD are fundamentally different. The metabolic component is characterized by hepatic oversupply of substrates, while the genetic component is characterized by impaired hepatic mitochondrial function.

Enhanced Palmitate-Induced Interleukin-8 Formation in Human Macrophages by Insulin or Prostaglandin E2

Janin Henkel et al.

Summary: The study found that a mix of palmitate, insulin, and PGE(2) in dysfunctional adipose tissue can induce the production of the pro-inflammatory cytokine IL-8. Insulin and PGE(2) amplify the effect of palmitate on IL-8 induction, exacerbating inflammation and insulin resistance.
