Topoisomerase I poison-triggered immune gene activation is markedly reduced in human small-cell lung cancers by impairment of the cGAS/STING pathway

Pharmacological Targeting of STING-Dependent IL-6 Production in Cancer Cells

Summary: Activation of the STING pathway in cancer cells upon genotoxic treatment leads to the production of interferon-beta and anti-tumoral effects. However, it is also associated with the expression of pro-tumorigenic genes. In this study, it was found that genotoxic DNA damage rapidly increased the expression of IL-6 in STING-expressing cancer cells, independent of type-I interferon responses. The production of IL-6 was driven through both canonical and non-canonical STING signaling pathways. Inhibition of ERK1/2 could inhibit IL-6 production induced by DNA damage.

Therapeutic targeting of ATR yields durable regressions in small cell lung cancers with high replication stress

Summary: Inhibiting ATR, the primary activator of replication stress, and TOP1, which suppresses genomic instability, synergistically showed cytotoxic effects in small cell lung cancer. The combination of ATR inhibitor M6620 and TOP1 inhibitor topotecan achieved a 36% objective response rate in SCLC patients, with durable tumor regressions observed in platinum-resistant SCNCs. These findings suggest that replication stress may be a transformative vulnerability in SCNCs, allowing for rational patient selection and potential personalized treatment strategies.
